补中益气汤对肺腺癌A549/DDP荷瘤小鼠LRP蛋白影响的研究

摘　　要：目的:探讨补中益气汤7、14、21d对荷瘤小鼠肿瘤中肺耐药蛋白(LRP)表达的影响。方法:体外培养A549/DDP细胞,接种健康BALB/c裸鼠,建立荷瘤裸鼠模型,随机分为:A549/DDP荷瘤对照组、DDP组、补中益气汤组及A549荷瘤对照组,观察并计算抑瘤率,并在7、14、21d脱颈处死小鼠,分别利用RT-PCR检测LRP mRNA的表达,免疫组化和Western Blot法检测中LRP蛋白表达。结果:补中益气汤能够抑制肺腺癌A549/DDP的生长,抑瘤率较A549/DDP荷瘤对照组相比明显增高,补中益气汤能在基因和蛋白层次上抑制LRP表达,且14、21d LRP下降程度逐渐增加。结论:补中益气汤能有效降低该实体瘤中LRP蛋白的表达。Objective： To study the effects of Buzhong Yiqi Decoction on the expression of lung resistance protein （LRP） in cells from A549/DDP tumor-bearing mice. Methods： A549/DDP cells were cultured in vitriol, and A549/DDP tumor-bearing nude mice model was established by inoculating healthy BALB/c nude mice with the A549/DDP cells. Tumor inhibitory rate was calculated after the A549/DDP tumor-bearing nude mice were treated with Buzhong Yiqi Decoction. All of the experimental mice were grouped into A549/DDP tumor control group, DDP group, Buzhong Yiqi Decoction group, and A549 tumor control group, and the mice were killed at different time points. Reverse transcliption-polymerase chain reaction （RT-PCR） was applied to determine the mRNA expressions of LRP mRNA, Western Blot and immunocytochemical method was adopted to determine the protein expression of LRP. Results： The growth of lung adenocarcinoma A549/DDP cells were inhibited by the Buzhong Yiqi Decoction, and the tumor inhibitory rate was significantly higher compared with the tumor control group. The expression of LRP was inhibited both in gene and protein level after the treatment of Buzhong Yiqi Decoction, and the effect became more significant with the lapse of time. Conclusion： Buzhong Yiqi Decoction serves effectively in decreasing the LRP protein expression of the tumor.
