摘 要:目的探讨结肠癌转移相关因子1(MACC1)和肝细胞生长因子(HGF)的表达与卵巢恶性上皮性肿瘤临床、病理特征的关系。方法免疫组织化学法和逆转录-聚合酶链反应(RT-PCR)检测47例上皮性卵巢癌、25例良性卵巢上皮性肿瘤、20例正常卵巢上皮性组织中MACC1、HGF蛋白及二者mRNA的表达。结果 (1)卵巢上皮性癌组织中MACC1和HGF蛋白的阳性表达率及相对表达量均显著高于良性上皮性肿瘤和正常卵巢组织(P均<0.05),而在卵巢良性肿瘤组织与正常卵巢组织中的表达差异无统计学意义(P>0.05);(2)卵巢上皮性癌组织中MACC1及HGF蛋白的表达与患者年龄、组织学类型无关(P均>0.05),与临床分期、组织学分级及淋巴结转移有关(P均<0.05);(3)MACC1 mRNA、HGF mRNA在卵巢上皮性癌组织中的表达量显著高于良性卵巢上皮性肿瘤和正常卵巢组织(F=295.032,P=0.00;F=177.252,P=0.00),且卵巢上皮性癌组织中MACC1 mRNA、HGF mRNA的表达水平呈正相关关系(r=0.810,P<0.01)。结论 MACC1可能在卵巢癌的恶性进展中发挥重要作用,有望成为上皮性卵巢癌诊断和治疗的靶点,与HGF联合检测对判断卵巢上皮性肿瘤预后有重要意义。Objective To investigate the relationship between the expressions of metastasis-associated in colon cancer 1 (MACC1), bepatoeyte growth factor (HGF) and the clinicopathological features of epithelial ovarian carcinoma. Methods Forty-seven patients with epithelial ovarian carcinoma, 25 patients with benign epithelial ovarian tumor and 20 patients undergone ovariectomy of normal ovarian duo to other reason were enrolled in present study. The expressions of MACC1 protein, HGF protein and MACC1 mRNA, HGF mRNA were detected by immunohistochemical method and reverse transcription-polymerase chain reaction (RT-PCR), respectively. Results ( 1 ) The positive expression rates and relative expression quantities of MACC1 protein and HGF protein in ovarian epithelial malignant tumor tissues were significantly higher than those in ovarian epithelial benign tumor tissues and normal ovarian epithelial tissues ( all P 〈 0. 05 ), and there is no significant difference between ovarian epithelial benign tumor tissues and normal ovarian epithelial tissues ( P 〉 0. 05 ). (2) The expressions of MACC 1 protein and HGF protein in ovarian epithelial malignant tumor tissues had no association with age of patient and histological type ( all P 〉 0. 05 ), but had obvious relation to clinical stage, histological grade and lymph node metastasis ( all P 〈 0. 05 ). (3) Compared with ovarian epithelial benign tumor tissues and normal ovarian epithelial tissues, the expression quantities of MACC1 mRNA, HGF mRNA in ovarian epithelial malignant tumor tissues increased significantly ( F = 295. 032 ,P = 0. 00 ; F = 177. 252, P = 0.00 ), and there was positive correlation between MACC1 mRNA and HGF mRNA expression levels in ovarian epithelial malignant tumor tissues ( r = 0. 810, P 〈 0.01 ). Conclusions MACC1 might play an important role in malignant progression of ovarian carcinoma, and it can be served as promising target for diagnosis and therapy of epithelial ovarian carcinoma.
