寒凝血瘀型大鼠胃黏膜内皮素-1与缺氧诱导因子-1α改变及对微循环血流量影响

摘　　要：目的:观察寒凝血瘀型大鼠胃黏膜内皮素-1(ET-1)与缺氧诱导因子-1α(HIF-1α)改变及对胃黏膜微循环血流量影响。方法:健康雄性Wistar大鼠20只,随机分为正常组与模型组(n=10)。模型组以冷暴露法模拟寒区低温(-10±2)℃,处理5 d,制造寒凝血瘀证大鼠模型,以免疫组化方法检测胃黏膜HIF-1α与ET-1表达,同时观察胃黏膜微循环血流量及血液流变学各项指标。结果:实验组冷暴露后出现寒战、爪蜷缩少动、尾部紫黯等症状,肉眼可见胃黏膜充血水肿与糜烂,光镜下可见有明显损伤;与正常组比较,HIF-1α与ET-1表达明显高于对照组(P<0.01);同时,胃黏膜微循环血流量明显少于正常组,血液流变学指标亦较正常组升高(P<0.01)。结论:冷暴露所致的大鼠寒凝血瘀证,可使其胃黏膜内皮素-1(ET-1)与缺氧诱导因子-1α(HIF-1α)表达增加,进而导致黏膜微循环血流量明显减少与血液流变学指标异常,最终造成胃黏膜损伤。Objective ： To observe the expresion of endothelin-1 （ ET-1 ） and hypoxia inducible factor- 1α （ HIF-1α ） in gastric mucosa of rats with blood stasis due to cold accumulation （ BTCA ） and effect on the blood flow and hemorheolog. Methods ： 20 Wistar rats were randomly divided into model group and normal control group （ n=10 respectively ）. The model group was exposed to cold （ -10 ± 2 ） ℃ for 5 days to make BTCA model and the normal control group was raised in room temperature and the expresions of ET-1 and HIF- 1 α and mucosal blood flow and hemorheology in gastric mucasa were observed. Result： Abnormalities were not observed in normal group rats. Rats in model goup appeared chill, hunched, lazy and cyanosis in tail and claw, and the injuries were observed in tunica mucosa both under naked eye and light microscope and its expressions of ET-1 and HIF-1α in gastric mucosa were higher than that in normal control group （P〈0.05）. Meanwhile, its hemorheology was increased remarkably and gastric mucosal blood flow was decreased obviously （ P〈0.01 ）. Conclusion ： The rats with BTCA displays high expressions of ET-1 and HIF-1 α in gastric mucosa what follows decreased gastric mucosal blood flow and increased hemorheology, finally causing the injury of mucous membrane.
