摘 要:目的:探讨低剂量长期连续天然放射性照射人群的热休克蛋白27(hsp27)基因表达水平。方法随机选择我国阳江市天然放射性高本底辐射地区( HBRA)50名50~59岁男性居民为HBRA组,另选择在恩平市某镇对照地区( CA)年龄相仿的50名男性居民为CA组。采集周围血分离血浆和白细胞,酶联免疫吸附实验测定血浆中HSP27蛋白水平,实时定量基因扩增荧光检测系统测定白细胞中hsp27 mRNA相对表达水平(以ΔCt值表示,ΔCt值越大表示mRNA相对表达水平越低)。数据采用中位数( M)与四分位数( P25,P75)描述。结果 HBRA组和CA组人群的血浆HSP27蛋白水平比较,差异无统计学意义[1.48(0.98,2.82) vs 1.48(1.12,2.45) mg/L,P >0.05]。 HBRA组周围血白细胞中hsp27 mRNA的ΔCt值低于CA组[1.68(1.33,2.35) vs 3.08(2.13,4.47),P<0.01],即HBRA组hsp27 mRNA相对表达水平高于CA组。结论低剂量长期连续照射可能提高人体白细胞hsp27基因表达水平,诱导产生适应性反应。Objective To examine the expression level of gene heat shock protein 27 (hsp27) among the residents chroni-cally exposed to low-dose continuous terrestrial radiation .Methods Fifty residents aged from 50 to 59 years old in Yangjiang high background radiation area ( HBRA) were randomly selected as the HBRA group and the other 50 residents in neighbo-ring Enping control area (CA) as the CA group.Peripheral blood was drawn from each subject and centrifuged to separate the plasma and white blood cells .The expression level of protein HSP 27 in the plasma was determined with enzyme-linked immunosorbent assay .The mRNA relative expression level of gene hsp27 in the white cells was described by ΔCT value which were determined with the real-time quantitative polymerase chain reaction detecting system , and the higher ΔCT value meant lower mRNA relative expression level .Median ( P25 , P75 ) was used to describe data .Results There was no significant difference found in plasma HSP27 level between the HBRA and CA groups [1.48 (0.98, 2.82) vs 1.48 (1.12, 2.45) mg/L, P>0.05 ] .The ΔCt value of gene hsp27 was significantly lower in the HBRA group than that in the CA group [1.68(1.33, 2.35) vs 3.08(2.13,4.47), P<0.01].It meant that the mRNA expression level of gene hsp27 was higher significantly in the HBRA group than that in the CA group (P<0.01).Conclusion It is inferred that chronic exposure to low-dose radiation may increase the expression level of gene hsp27 in the white cells and induce adaptive response .
