摘  要:将虹鳟鱼苗研磨过滤除菌后接种到胖头鱼肌肉细胞系(FHM)后出现了特征性病变(CPE),电镜下观察到IHNV病毒粒子。用传染性造血器官坏死病病毒(IHNV)和传染性胰腺坏死病病毒(IPNV)等水生动物病毒特异性引物对该毒株进行RTPCR试验,结果显示,用IHNV的引物能扩增出阳性片段。将分离株暂时命名为CJ-13。应用DNAStar和MEGA5.2软件,将CJ13的N基因与多株GenBank中已发表的IHNV毒株相应基因进行同源性比对和构建系统树。核苷酸同源性显示CJ-13株与HV7601株的N基因同源性高达97.1%,推导出的氨基酸序列同源性为95.2%。系统进化树表明,CJ-13分离株与HV7601株遗传进化关系较近,属于同一分支。A strain of rhabdovirus was isolated from the larvae of rainbow trout.The obvious cytopathic effect (CPE) was observed in Fathead minnow (FHM) inoculated with tissue homogenates and IHNV particles were observed under electron microscope. Reverse transcription polymerase chain reaetion(RT-PCR) was tested with specific primers of infectious hematopoietic necrosis virus (IHNV), infectious pancreatic necrosis virus (IPNV) and other aquatic animal virus.The fragment was amplified with the primers of IHNV and was assigned C J-13.The sequence of the N gene of C J-13 was compared with the sequences of other IHNV strains in GenBank by DNAStar and MEGA5.2 software.Sequence analysis showed that the nucleotide sequence similarity with HV7601 was 97.1%, while the deduced amino acid sequence similarity was 95.2%.The two strains C J-13 and HV7601had the closest genetic rela- tionship and belonged to the same branch.
