摘 要:目的:分析SHP-1基因和JAK家族基因在成人急性髓系白血病(AML)患者中的表达及相关性,以探讨两者在AML发病机制中的作用。方法:选取初发急性髓系白血病患者93例及健康志愿者或完全缓解(CR)患者20例为研究对象,分别应用荧光定量聚合酶链(FQ-PCR)和反转录聚合酶链反应(RT-PCR)方法检测骨髓单个核细胞中SHP-1和JAK3基因的mRNA表达情况,并分析两者的相关性及其与预后的关系。结果:SHP-1在AML患者中的表达水平明显低于完全缓解组和健康对照组(P<0.01),而JAK3表达水平明显高于完全缓解组和健康对照组(P<0.05);SHP-1和JAK3在mRNA水平方面呈现负相关(P<0.05)。结论:SHP-1可能是白血病的抑癌基因,AML细胞中SHP-1与JAK3的mRNA表达呈负相关,这种负调控作用的机制有待进一步研究。Objective:To investigate the expression of SHP - 1 and JAK3 mRNA in the primary acute myloid leukemia (AML) patients and their impact on leukemogenesis. Methods:The expression of SHP - 1 and JAK3 mRNA in 93 AML pa- tients and 20 healthy or complete remission(CR) adults as normal controls were measured with fluoreseenee quantitative poly- merase chain reaction (FQ -PCR) and reverse transcription polymerase chain reaction( RT -PCR). The relationship between the expressions of SHP - 1 and JAK3 was analysed. Results : The expression of SHP - 1 mRNA in de novo AML patients was significantly lower than that in complete remission and healthy control group( P 〈 0.01 ). The expressions of JAK3 in AML were significant higher than complete remission and healthy control group ( P 〈 0.05 ). There was a negative correlation between SHP - 1 and JAK3 mRNA ( P 〈 0.05 ). Conclusion : SHP - 1 gene is a antioncogene. There was a negative correlation between SHP - 1 and JAK3 mRNA. Such negative regulation mechanism needs further study.
