摘 要:目的观察活血益智片对脑血管性痴呆(VD)大鼠脑组织N-甲基D-天冬氨酸受体(NMDAR)表达及一氧化氮(NO)5-羟色胺(5-HT)含量影响。方法反复缺血再灌注建立VD大鼠模型,用活血益智片灌胃给药28 d后,采用Morris水迷宫进行大鼠学习记忆能力测试,免疫组化法观察大鼠脑组织中NMDAR表达;用液质联用仪测定大鼠脑组织中5-HT含量;硝酸还原酶法测定NO含量。结果与对照组比较,模型组大鼠逃避潜伏期[(34.11±13.58)s]明显延长,脑内5-HT含量[(20.26±0.57)ng/g]下降,NO含量[(132.61±33.99)μmol/gprot]明显升高,脑组织NMDAR表达增加;与模型组比较,6.20 g/kg活血益智片组大鼠逃避潜伏期[(15.18±6.82)s]明显缩短,脑内5-HT含量[(26.23±3.08)ng/g]增加,NO含量[(95.57±29.06)μmol/gprot]下降(P<0.05),脑组织NMDAR表达减少。结论活血益智片可明显改善血管性痴呆大鼠学习记忆能力,其机制可能与增加大鼠脑组织中5-HT含量、减少NMDAR表达有关。Objective To observe the effect of Huoxue-Yizhi tablet on brain N-methyl-D-aspatric acid receptor (NMDAR) expression and the contents of nitric oxide(NO) and 5-hydroxy trypotamince(5-HT) ,and to explore protec- tive mechanism of the tablet. Methods Repeated ischemia reperfusion was used to establish vascular dementia (VD) model in rats with oral administration of Huoxue-Yizhi tablet for 28 days. Learning and memory ability of the rats were tested using Morris water maze. The expression of NMDAR in brain tissue was determined with immunohistochemistry; 5-HT content in cerebral tissue was measured with liquid chromatography-mass spectrometry and NO content was determined with nitrate reductase method. Results Compared with the control group, the escape latency ( 34. 11 ± 13.58 s) in model group was significantly longer; the content of 5-HT (20. 26 ± 0. 57 ng/g) was decreased and the NO content( 132. 61 ± 33.99 μmol/gprot) was increased;the expression of NMDAR was downregulated. Compared with the model group ,the escape latercy( 15.18 ± 6. 82 s) was significantly shortened;the content of brain 5-HT(26. 23 ±3. 08 ng/g) was increased;and NO content( 95.57 ± 29. 06 μmol/gprot) and NMADR expression was decreased( P 〈 0. 05 ) in the rats with high-dose (6.20 g/kg) Huoxue-Yizhi tablet treatment. Conclusion Huoxue-Yizhi tablet can obviously improve the behavior of vascular dementia rats, and the effect might be related to the increased NO and 5-HT and decreased NMDAR expression in the brain of the rats.
