摘 要:目的:下调胃癌细胞HSP27的表达水平,观察胃癌细胞侵袭转移能力的变化。方法:利用Western blot方法选取HSP27高表达的胃癌细胞系,采用siRNA技术下调HSP27的表达,通过Transwell小室和划痕实验观察细胞系侵袭转移能力的变化。结果:Western blot实验结果显示,BGC823细胞和MKN28细胞中HSP27的呈高表达状态。转染HSP27-siRNA后,BGC823细胞和MKN28细胞中HSP27在蛋白水平和mRNA水平表达量均有显著地下降,验证转染成功。Transwell小室和划痕实验发现,转染HSP27-siRNA后BGC823和MKN28细胞的侵袭转移能力显著低于阴性对照组。结论:HSP27与胃癌细胞的侵袭转移能力密切相关,抑制HSP27的表达能够使胃癌细胞的侵袭转移能力明显下降。Objective:To analyze the relationship between the expression d heat shock protein 27 (HSP27) and the invasive and metastatic potential of human gastric cancer (GC) cell lines. Methods:Western blot was used to choose cell lines which highly expressed HSP27. Lentivirus mediated siRNA technique was performed to down - regu- late the expression of HSP27 in BGC823 and MKN28 gastric cancer cell lines. Western blot and RT - PCR were em- ployed to test the infection efficiency. Transwell and scratch assay were used to identify the changes of invasive and metastatic ability in BGC823 and MKN28 gastric cancer cell lines. Results:Western blot showed that expression level of HSP27 in BGC823 and MKN28 cells were relatively high among 6 gastric cancer cell lines. The invasive and meta- static potentials of BGC823 - si and MKN28 - si were significantly decreased after down - regulating HSP27. Conclu- sion:The expression level of HSP27 may be correlated to the invasive and metastatic potential of GC. HSP27 may play an important role in the invasion and metastasis of GC cell lines. HSP27 may be a potentiul molecular target for the di- agnosis,therapy and prognosis of GC patients.
