摘 要:目的探讨O^6-甲基鸟嘌呤-DNA甲基转移酶(MGMT)基因rs1625649多态性与乳腺癌易感性的关系。方法乳腺癌患者55例,健康对照者119例,采用高分辨熔解曲线法(HRM)进行突变筛查,并进行测序验证。结果乳腺癌患者MGMT基因第2启动子区域检测到1个多态性位点,经测序验证,该位点为rs1625649。MGMTrs1625649位点A携带患者的危险度较对照组升高[P〈0.05,比值比(OR)=1.31,95%可信区间(CI):1.09—1.59]。基因型分析结果表明MGMTrs1625649位点的AA基因型是乳腺癌高危基因型[P〈0.05,OR=1.46,95%CI:1.06~2.01]。结论MGMTrs1625649的基因多态性可能与乳腺癌的发生发展有关。Objective To expolre the relationshio between genetic polymorphisms of O"-methyl- guanine-DNA methytransferase (MGMT) and susceptibility of breast cancer. Methods Totally 55 breast cancer patients and 119 non-cancer female controls were selected. The genotypes were detected by using high resolution melting curve method (HRM) and verified by sequencing. Results The risk of breast cancer was increased in MGMT rs1625649 A genetic carrier [ P 〈 0. 05, odds ratio (OR) = 1.31, 95% confidence interval (CI) = 1.09-1.59 ] as compared with the controls. MGMT rs1625649 AA was the high risk genotype ( P 〈 0. 05, OR = 1.46, 95 % CI = 1.06-2. 01 ). Conclusion The results indicated that the polymorphisms in MGMT genes might be associated with breast cancer risk of Chinese women.
