β-榄香烯联合顺铂对乳腺癌MCF-7细胞增殖和凋亡的影响

摘　　要：目的:探讨β-榄香烯联合顺铂对乳腺癌MCF-7细胞生长的影响。方法:体外培养乳腺癌MCF-7细胞,将β-榄香烯(浓度梯度为25、50、100、150、200μg/ml),单独作用于乳腺癌MCF-7细胞,加药24h、48h后用噻唑蓝(MTT法)法检测细胞增殖情况。用流式细胞术检测用药24h后对MCF-7细胞凋亡和细胞增殖周期的影响,选取合适的药物浓度(β-榄香烯125μg/ml),与顺铂(3μg/ml)进行联合用药。加药24h、48h用MTT法检测细胞增殖情况,用流式细胞术检测24h后药物组对MCF-7细胞凋亡和细胞增殖周期的影响。结果:MTT法结果显示β-榄香烯单独用药24h、48h后,与对照组相比,实验组乳腺癌MCF-7细胞的抑制率高于对照组(P<0.05),并且在一定程度上呈浓度和时间依赖性。联合用药时,细胞的抑制率和凋亡率要显著高于单独用药(P<0.01)。与对照组相比,榄香烯能够使MCF-7细胞产生明显的G0/G1期阻滞(P<0.01)。结论:β-榄香烯单独或与顺铂联合作用均能抑制MCF-7细胞的增殖,促进其凋亡,且β-榄香烯联合顺铂的作用要显著高于单独用药组,β-榄香烯和顺铂可协同(CDI<1)促进MCF-7细胞凋亡,其机制可能与G0/G1期阻滞有关。Objective：To investigate the effect of β - Elemene combined with DDP on the Proliferation and aPoP-tosis of MCF - 7 cell. Methods：Different concentrations of β - Elemene injection（25,50,100,150,200μg/ ml）were aPPlied to MCF - 7 cells. Cell Proliferation was measured by MTT. Cell aPoPtosis and cell - cycle was detected by flow cytometry. The aPProPriate drug concentration（3μg/ ml DDP,125μg/ ml β - Elemene）were selected. Then β - El-emene combined with DDP injection was aPPlied to MTT cells. Cell Proliferation was measured by MTT. Cell aPoPtosis and cell - cycle was detected by flow cytometry. Results：Cell Proliferation was dramatically inhibited by β - Elemene injection（25,50,100,150,200μg/ ml）at 24h,48h（P ﹤ 0. 05）. In addition,cell Proliferation was dramatically inhib-ited by injection β - Elemene combined with DDP. Combination theraPy,aPoPtosis rate and inhibition rate of Prolifera-tion in MCF - 7 cells treated by β - Elemene combined with DDP was higher than the monotheraPy（P ﹤ 0. 01）. Cell- cycle was arrested in G0 / G1 Phase by elemene comPared to uninfectious cells in the same time scale. Conclusion：β - Elemene and DDP combined with β - Elemene can inhibit the Proliferation,accelerate the aPoPtosis in MCF - 7 cells. DDP combined with β - Elemene have synergistic effect on the aPoPtosis in MCF - 7 cells.
