摘  要:目的探讨miR-31在宫颈癌组织中的表达,并对其进行靶基因功能预测等生物信息学分析,为miR-31在宫颈癌发病机制中作用的研究提供理论和实验基础。方法应用实时荧光定量PCR技术检测宫颈癌组织及正常宫颈组织中miR-31相对表达量;通过mirecord网站对miR-31进行靶基因预测,并利用DAVID对靶基因进行GO(gene ontology)功能富集分析及KEGG信号转导通路富集分析。结果宫颈癌组织中miR-31表达(△Ct=4.53±1.64)水平高于正常宫颈组织(△Ct=7.46±1.38);miR-31预测靶基因集合功能富集于生物学调控、蛋白代谢、细胞过程,有机体的发育等生物学过程,以及蛋白结合、底物特异性的跨膜转运活动等分子功能上;KEGG通路分析涉及癌症通路、黏着连接、内吞作用、黑色素瘤、长时程增强、轴突导向等。结论 miR-31在宫颈癌组织中呈高表达,miR-31预测靶基因集合显著富集在与肿瘤发生相关的信号通路中。Objective To investigate the expression of miR-31 in cervical cancer tissues and analyze bioinformatically the target genes of miR-31 in order to provide theoretical basis for further studies of miR-31 biological function in cervical cancer. Methods Real-time PCR was used to access the expression of miR-31 in cervical tissues and normal cervical tissues, and mirecord was used to predict target genes of miR-31. DAVID was used to analyze the GO (gene ontology) function annotation and the KEGG signal pathway of the miR-31 target genes. Results The expression of miR-31 was significantly higher in cervical cancer tissues (△Ct= 4.53 ±1.64) than that in normal cervical tissues (△Ct= 7.46±1.38). The function of miR-31 for predicting target genes was enriched in biological regulation, protein metabolism, cellular process, developing process, protein binding, substrate-specific transmembrance transporter activity, and other biological processes and molecular functions. In KEGG pathway, the predicted target genes involved in pathway in cancer, adherence junction, endocytosis, melanogenesis, long-term potentiation and axon guidance. Conclusions The expression of miR-31 is significantly high in cervical cancer tissues. The predicted target genes of miR-31 are significantly enriched in tumor related signaling pathways.
