摘 要:目的考察TGF-β1对人粘连成纤维细胞(ATF)增殖与COL-Ⅰ合成的影响。方法通过人体腹膜粘连组织原代培养获取ATF,将不同剂量的TGF-β1与ATF共培养48h,MTT法测定细胞增殖活性,ELISA法测定培养基中COL-Ⅰ的含量。结果TGF-β1 0.5~40ng/mL剂量组的ATF增殖活性均高于0ng/mL剂量组,差异有统计学意义(P〈0.01)。TGF-β1 10、20ng/mL剂量组的COL-Ⅰ含量与0ng/mL剂量组差异有统计学意义(P〈0.01)。结论TGF-β1可明显提高人ATF的增殖活性与COL-Ⅰ合成水平。Abstract: Objective To study the effect of TGF-β1 on the proliferation and COL-I synthesis of human fibroblasts from adhesive tissue (ATF). Methods ATF from human peritoneal adhesive tissues were harvested by primary culture. The proliferation of ATF treated with different dosage of TGF-β1 was measured by MTT assay. ELISA was employed to determine the content of COL-Ⅰ in ATF culture medium. Results Compared with the control group, ATF treated with the dosage from 0.5 to 40 ng/mL TGF-β1 showed an higher viability (P〈O.01). Meantime, COL-I content was significantly increased when ATF treated with 10 and 20 ng/mL TGF-β1 (P〈0.01). Conclusion TGF-β1 may increase the proliferation and COL-Ⅰ synthesis of human ATF.
