摘 要:目的建立一种基于Kligman痤疮模型的痤疮复合动物模型,以期获得更接近临床症状的痤疮模型。方法将家兔随机分为正常组、注射病菌组、Kligman组和复合模型组。正常组不做处理,Kligman组和复合模型组均在家兔耳内侧涂抹2%煤焦油持续2周,注射病菌组及复合模型组在造模期间隔日进行痤疮丙酸杆菌(Propionibacterium acnes)液的接种,出现可见囊肿后即可停止接种。模型制备成功后,测量各组表面微循环,检测耳增厚程度,并进行病理组织检查。采用酶联免疫吸附测定法(Elisa)检测血清中白细胞介素-1α(IL-1α)、白细胞介素-6(IL-6)和二氢睾酮(DHT)浓度。结果复合模型组与Kligman组比较,IL-1α差异无统计学意义(P>0.05),IL-6增加显著(P<0.01),DHT有一定程度的上升(P<0.05);复合模型组较Kligman组兔耳肿胀程度显著性增加(P<0.01),血液微循环差异有统计学意义(P<0.05)。病理组织观察显示复合模型组表皮角化过度增厚,颗粒层、棘层肥厚,毛囊口充满角化物质,毛囊口由于过多的内容物而膨胀突出,毛囊漏斗部破裂,可见炎症细胞浸润。痤疮复合模型相对于Kligman模型表现出更严重的痤疮反应。结论该方法能制备符合临床症状的痤疮复合模型。Objective To establish a acne complex animal model based on the Kligman model close to clinical symptoms.Methods Coal tar was smeared on the inside of New Zealand Rabbit's auricle for two weeks and then P. ache was injected to build the ache complex animal model.The local microcirculation, ear slice weight, and the concentration of DHT, IL-1α and IL-6 in the serum were measured.Results Compared to the Kligman model, the level of IL-1α was no significant difference(P〉0.05) ,but the weight of ear slice (P〈0.01), the concentration of IL-6 (P〈0.01) and the concentration of DHT (P〈0.05) increased in the complex model group.Meantime, the local mierocircnlation was significant difference (P〈0.05) between two groups.Compared with the Kligman model,epidermis hyperkeratosis, granular layer and stratum spinosum thickened, hair follicles filled with the keratin,infundibulum rupture of hair follicle and inflammatory cells infiltration were more apparent in the complex model group.Conclusion This new method may be more close to the clinical symptoms of acne.
