摘 要:microRNAs是一类单链、小分子非编码RNAs,通过调控基因表达,对于生命活动的各个过程起着重要的调控作用。通过作用于相关的mRNA分子,microRNAs调控肿瘤的发生和发展。microRNAs对于基因表达表达的发挥重要的调控作用,到底对肿瘤的发生具有促进作用还是抑制作用一直存在争议。到目前为止,作为肿瘤标记物和治疗的靶向分子还存在很大的限制性。文中涉及microRNAs的正负功能态调节作用,从理论上阐明其在肿瘤转移中的作用,并对microRNA的临床应用及抗癌前景提出了展望。MicroRNAs are single-stranded RNA molecules that control gene expression in many cellular processes. These molecules typically reduce the translation and stability of mRNAs, including those genes that mediate processes in tumorigenesis. MicroRNAs have been established as key regulators of gene-expression, and their putative roles as oncogencs and turnout suppressor genes has provided a potentially new dimension to our clinical approach to cancer diagnosis and treatment. Their role as biomarkers and therapeutic targets is appealing but several obstacles have as yet limited our ability to translate this potential into a clinical reality. This review focuses on currently accepted roles of microRNAs in cancer pathogenesis, and highlights the challenges and breakthroughs in this field to date with relevance to the cancer clinic.
