摘  要:目的研究芦丁对大鼠心肌缺血再灌注损伤的保护作用及其机制。方法通过左冠状动脉结扎建立SD大鼠心肌缺血再灌注模型,将大鼠随机分为对照组、模型组、芦丁组、L-NAME(eNOS抑制剂)组,ELISA法检测血清一氧化氮(NO)水平,免疫抑制法检测血清肌酸激酶同工酶(CK-MB)浓度,实时荧光定量PCR分析各组心肌组织iNOS和eNOS mRNA表达的变化。结果与模型组比较,芦丁组血清NO水平增高(P<0.01),CK-MB浓度下降(P<0.01),eNOS mRNA表达升高(P<0.01),iNOS mRNA表达下降(P<0.01)。结论芦丁缺血预处理对心肌起到了保护作用,其机制可能与上调心肌组织eNOS mRNA、下调iNOS mRNA表达有关。Objective To investigate the protective effect of Rutin on myocardial tissue of rats with ischemia-reperfusion injury.Methods Rat model of ischemia-reperfusion injury was set up by ligating the left anterior de-scending batch of the coronary artery.SD rats were randomly divided into control group ,model group ,Rutin group and L-NAME (eNOS inhibitor) group.Serum level of NO was determined by ELISA and CK-MB concentration was examined by immun osuppression assay.The expression of iNOS and eNOS mRNA in myocardial tissues was evaluated by real-time quantitative PCR analysis.Results Compared with model group ,serum level of NO was in-creased while CK-MB concentration was significantly decreased in rats of Rutin group (P 〈0.01).The expression of eNOS mRNA was up-regulated while the expression of iNOS mRNA was down-regulated in myocardial tissues of rats in Rutin group (P〈0.01).Conclusion Ischemic preconditioning with Rutin is effective in the treatment of ischemia-reperfusion injury.The mechanism might be associated with regulating the expressions of eNOS and iNOS mRNA in myocardial tissues.
