纤溶酶原激活物抑制剂-1及其基因启动子区4G/5G多态性在大肠癌与癌旁组织中的表达及意义

摘　　要：目的:探讨纤溶酶原激活物抑制剂-1(PAI-1)及其基因启动子区的一个鸟嘌呤插入/缺失多态性(4G/5G)与大肠癌转移的关系。方法:运用PCR技术和琼脂糖电泳对30例大肠癌患者癌变大肠组织与癌旁正常大肠组织进行PAI-l基因4G/5G多态性检测和分析,并将两者进行比较。采用酶联免疫吸附法检测上述标本组织PAI-1含量。结果:大肠癌患者PAI-1三种基因型在癌变大肠组织和癌旁正常大肠组织的表达比较,差异无统计学意义(P>0.05);组织PAI-1含量在大肠癌患者中高表达,与癌旁正常大肠组织比较,差异有统计学意义(P<0.05)。结论:PAI-1含量高表达与大肠癌的转移有关,但PAI-1基因多态性与大肠癌的转移无关。Objective： To investigate the plasminogen activator inhibitor -1 （PAI-1） and the promoter of a guanine sub region of the insertion/deletion polymorphism （4G/5G） relationship with the occurrence and development of colorectal cancer.Method： Using PCR technique and agarose gel electrophoresis of PAl-1 gene 4G / 5G polymorphism detection to analyze 30 cases of colorectal cancer colorectal cancer tissue and adjacent normal colorectal tissues, and compare them.Using enzyme-linked immunosorbent method to detect the contents of the specimen organization PAI-1.Result： No significant expression of PAI-1 of three gene in patients with colorectal cancer in colorectal cancer tissues and adjacent normal colorectal tissue differences（P〉0.05）; tissue PAI-1 content was high expressed in patients with colorectal cancer（P〈0.05）.Conclusion： The high expression of PAl-1 is related to the transfer of colorectal cancer, but PAI-1 gene polymorphism is not related to the transfer of colorectal cancer.
