摘  要:为检测细胞骨架抑制剂对狂犬病病毒(RABV)感染细胞的抑制作用,筛选了秋水仙素(Col)和细胞松弛素D(CytoD)2种骨架特异性抑制剂在人神经瘤细胞SH-SY5Y上的适宜浓度,探讨其对RABV感染及复制的影响。结果表明:随着细胞骨架抑制剂浓度的升高,微管和微丝的破坏加剧;当Col浓度分别为0.065 mmol/L和0.130 mmol/L及CytoD浓度分别为0.25μg/mL和0.50μg/mL时,微管、微丝结构及细胞活力均与对照组无显著差异(P>0.05),2种骨架抑制剂对RABV的感染及复制均有抑制作用。通过对RABV培养物的OD值和G基因mRNA的定量检测,发现CytoD对RABV感染的抑制作用较Col更明显,Col对RABV复制的抑制作用较CytoD更明显。To study the inhibitory effects of cytoskeleton inhibitors on rabies virus( RABV) infection in vitro,colchicine( Col) and cytochalasin D( CytoD) were chosen,their suitable concentration on neuroblastoma( SH-SY5Y) was screened,and their effects on RABV earlier infection and replication were evaluated. The results showed that aggravated damage of microtubules and actin filaments was closely associated with the highly elevated concentration of inhibitors. When Col was at 0. 065 mmol / L and 0. 13 mmol / L,respectively,and CytoD at 0. 25 μg / mL and 0. 50 μg / mL,no significant structural changes of microtubules and actin were observed compared with the control( P 〉 0. 05); the two cytoskeleton inhibitors demonstrated inhibitory ability in RABV infection and replication,RABV titer and Q-PCR of G gene mRNA confirmed that CytoD played a more important role in early RABV infection than Col,however,a more significant role of the Col was proved in RABV replication than CytoD. All these results provided experimental data for cytoskeleton associated RABV pathogenesis.
