摘  要:以猪整联蛋白α9(Integrin alpha 9,ITGA-9)基因为候选基因,通过相关分析发掘该基因与三元杂种猪滴水损失性状关联的分子遗传标记。采用PCR-SSCP和RT-PCR方法对98头洋三元杂种猪ITGA-9基因第3内含子进行多态性检测,分析其与滴水损失性状和ITGA-9基因表达水平的关系。结果表明:在ITGA-9基因第3内含子1791碱基处发现c.158+1731C>T突变,C和T等位基因频率分别为0.469和0.531。统计分析表明,CC型个体的滴水损失显著低于TT型个体(P<0.05),且CC型个体mRNA表达水平显著低于TT型个体。ITGA-9基因mRNA表达水平与相应的滴水损失性状之间存在正相关关系(r=0.459,P<0.05),且在滴水损失高低组间存在显著差异。ITGA-9基因c.158+1731C>T突变与猪肉滴水损失性状及ITGA-9基因表达水平相关。This study was to investigate the relationship of integrin alpha 9(ITGA-9) gene polymorphisms with drip loss trait in Duroc× Landrace×Yorkshire(D×L×Y) pigs. PCR-SSCP and DNA sequencing were employed to reveal the polymorphisms in intron 3 of ITGA-9 using a population of 98 D×L×Y pigs. The correlation of the novel mutation with drip loss traits was tested. The effect of the mutation on ITGA-9 mRNA expression level was evaluated as well. C〉T mutation harbored in intron 3 was identified at c. 158 +1731 site of ITGA-9 gene. The allele frequencies were 0. 469 for C and 0. 531 for T,respectively. Statistical analysis revealed that drip loss of CC genotypes is lower than that of TT(P〈0. 05). Correspondingly,CC genotype exhibited lower ITGA-9 mRNA level than TT genotype(P〈0. 05). The ITGA-9 mRNA level between corresponding traits of drip loss has positive correlation,and the higher drip loss group is significantly higher than the lower drip loss group(P 〈 0. 05). We identified c. 158 + 1731C 〉 T mutation of ITGA-9 gene to be correlated with ITGA-9 mRNA level and pork drip loss traits.
