摘 要:目的探讨缺氧条件下血管内皮生长因子受体2(vascular endothelial growth factor receptor 2,VEGFR2)对视网膜视锥细胞的作用。方法 8周龄视锥细胞特异性敲除VEGFR2(cone-specific VEGFR2 knockout)小鼠(简称KO小鼠)和野生型C57BL/6小鼠(简称WT小鼠)各6只,右眼均予以玻璃体腔内一次注射8 mmol/L氯化钴(CoCl2)制作缺氧模型,左眼注射磷酸盐缓冲液(PBS)作为对照。次日行视网膜电图(ERG)检查光感受器功能,免疫荧光共定位法检测视锥细胞密度,并观察其形态。结果 KO和WT小鼠缺氧侧眼的暗适应ERG a波振幅和b波振幅均下降,与PBS对照侧相比差异有统计学意义(P<0.01),但KO小鼠与WT小鼠之间相比差异无统计学意义(P>0.05)。KO小鼠和WT小鼠缺氧侧眼的明适应ERG a波振幅和b波振幅较对照侧均下降,差异有统计学意义(P<0.01),且KO小鼠较WT小鼠下降更明显(P<0.05,P<0.01),说明视锥细胞功能下降。形态学观察发现KO小鼠和WT小鼠缺氧侧眼的视锥细胞均出现变化,表现为密度下降,形态短小,排列疏松、紊乱,且KO小鼠的改变更为显著。结论 VEGFR2在缺氧条件下对视锥细胞具有保护作用。Objective To investigate the protective effect of VEGF receptor 2 (VEGFR2) on retinal cone photoreceptors under hypoxia condition. Methods Eight-week-old cone-specific VEGFR2-knockout (KO) mice and wild-type (WT) C57BL/6 mice received intravenous injection of 8 mmol/L cobalt chloride in the right eyes for hypoxia condition and PBS in the left eyes for control, with 6 mice in each group. On the next day electroretinogram (ERG) was conducted to evaluate the photoreceptor function, and immunofluorescence colocalization was used to determine the density and morphology of cone photoreceptors. Results In KO and WT groups, hypoxia eyes had significantly decreased a-wave and b-wave amplitudes in scotopic ERG compared with PBS control eyes (P〈0.01), but with no significant difference between hypoxia eyes of KO and WT groups (P〉0.05). In both KO and WT groups, the hypoxia eyes also had significantly decreased a-wave and b-wave amplitude in photopic ERG compared with PBS control eyes (P〈0.01), and the decrease in KO mice was more significant than that in WT group (P〈0.05, P〈0.01), indicating the decrease of cone function in KO animal. Morphology examination showed noticeable changes under hypoxia condition in both KO and WT groups, showing decreased density, shortened appearance, and disordered arrangement, with the changes in KO group being more noticeable. Conclusion VEGFR2 has a protective effect on cone photoreceptors under hypoxia condition.
