牙髓卟啉单胞菌密度感应系统luxS基因的克隆及测序

摘　　要：目的检测牙髓卟啉单胞菌模式株、临床分离株密度感应系统luxS基因。方法选取牙髓卟啉单胞菌模式株及临床分离株作为研究对象,提取细菌基因组DNA,PCR克隆及测序,并通过基因序列比对,比较临床分离株和模式株的一致性。结果电泳鉴定存在目的条带,克隆测序和序列比对表明,牙髓卟啉单胞菌PCR产物与目的基因完全一致(均为100%)。结论本实验引物设计合理,能较好地扩增出牙髓卟啉单胞菌的luxS基因,并对其进行测序,为进一步研究牙髓卟啉单胞菌luxS基因的功能奠定基础。Objective To detect the presence of luxS gene in quorum sensing signal system in Porphyromonas endodontalis （P. e）. Methods The total DNA of P. e was extracted. The presence of luxS was examined by PCR. The products of PCR were detected by electrophoresis, sequenced and identified by sequence alignment with the sequence in the Human Oral Microbiome Database （HOMD）. Results Electrophoresis, sequencing and sequence alignment indicated that the PCR products of P. e were highly consistent with the luxS gene in HOMD. Conclusion The designed primer is reasonable and luxS gene of P. e can be successfully amplified,which laid the foundation for further study on the function of luxS gene.
