摘　　要：目的:探讨水通道蛋白5(AQP5)在人宫颈癌中的表达情况及其临床意义。方法:分别通过qRT-PCR和Western blotting方法检测宫颈癌及癌旁组织中AQP5 mRNA和蛋白的表达情况,并分析AQP5 mRNA的表达与临床病理特点的关系。采用ROC曲线分析癌症患者组织中AQP5 mRNA的诊断价值。结果:AQP5 mRNA和蛋白水平在宫颈癌组织中的表达均高于癌旁正常组织(P<0.01),且AQP5的过表达与宫颈癌的淋巴结转移(P<0.01)及肿瘤分期(P<0.05)有关。ROC曲线结果显示AQP5在宫颈癌组织中的检测敏感性、特异性及AUC值可分别达到72.2%、75.0%和0.715。结论:AQP5在宫颈癌的发生发展中起着重要作用,有机会成为新的宫颈癌的诊断标志物及治疗靶点。Objective： To explore the expression of aquaporin 5（ AQP5） in human cervical cancer and its clinical significance.Methods： qRT- PCR and Western blotting were used to detect the expressions AQP5 mRNA and protein in cervical cancer tissue and para- carcinoma tissue; the relationship between AQP5 mRNA expression and clinicopathological features was analyzed; the diagnosis value of AQP5 mRNA in patients with cervical cancer was analyzed by ROC curve analysis. Results： The expressions of AQP5 mRNA and protein in cervical cancer tissue were statistically significantly higher than those in normal para- carcinoma tissue（ P〈0. 01）. Over- expression of AQP5 was significantly correlated with lymph node metastasis（ P〈0. 01） and neoplasm staging（ P〈0. 05）. The results of ROC curve showed that the sensitivity,specificity and AUC value of AQP5 in cervical cancer tissue were 72. 2%,75. 0% and 0. 715,respectively.Conclusion： AQP5 plays a key role in tumorigenesis and development of cervical cancer,which may become to be a new diagnosis marker and therapeutic target of cervical cancer.
