摘 要:目的探讨短期乙醇暴露后雌性大鼠肝功能酶类指标的动态变化。方法将40只健康成年雌性大鼠随机分为对照(无菌蒸馏水)组和低剂量(7.893 g/L)、中剂量(15.786 g/L)、高剂量(23.679 g/L)乙醇染毒组,每组10只。采用腹腔注射方式进行染毒,染毒量为5 ml/kg,1次/d,连续染毒4周。检测大鼠血清碱性磷酸酶(ALP)、乳酸脱氢酶(LDH)、谷氨酸氨基转移酶(ALT)、天门冬氨酸氨基转移酶(AST)和胆碱酯酶(CHE)活性。结果随着乙醇暴露时间的延长,不同浓度乙醇组大鼠血清ALT活性均呈上升趋势(P<0.05);不同浓度乙醇组血清AST活性均呈先升高后降低的趋势(P<0.05);血清LDH活性随着乙醇暴露时间的延长呈先升高后降低的趋势,在乙醇暴露第4周时,高剂量组血清LDH活性明显低于对照组(P<0.05);随着乙醇暴露时间的延长,对照组和低剂量组、中剂量组血清ALP活性均呈下降趋势,而高剂量组血清ALP活性呈先降低后升高的趋势,在乙醇暴露第4周时,高剂量组血清ALP活性明显高于对照组(P<0.05);随着乙醇暴露时间的延长,对照组、低剂量组、中剂量组血清CHE活性均呈上升趋势,而高剂量组血清CHE活性则呈下降趋势,在乙醇暴露后第2、4周时,高剂量组血清CHE活性均明显低于对照组(P<0.05)。结论乙醇暴露导致雌性大鼠肝损伤,其敏感血清酶指标为CHE、LDH及ALP,且CHE更敏感,均呈动态变化。Objective To investigate the dynamic changes of liver function enzymes of short-term ethanol-exposed female rats. Methods Forty healthy adult female rats were randomly divided into control group,low-dose ethanol group,middle-dose ethanol group and high-dose ethanol group,with 10 rats in each group. The rats in the control group were given sterile distilled water,while in the low-,middle-and high-dose ethanol groups were given 7. 893 g / L,15. 786 g / L and23. 679 g / L ethanol,respectively. The mice were treated by intraperitoneal injection,with the dose of 5 ml / kg,once a day for 4 weeks. The activities of alkaline phosphatase( ALP),lactic dehydrogenase( LDH),glutamic-pyruvic transaminase( ALT),glutamic-oxalacetic transaminease( AST) and cholinesterase( CHE) in serum were detected. Results With the increasing time of ethanol exposure,the activity of serum ALT increased( P〈0. 05),the activity of serum AST increased and then decreased in different doses ethanol groups( P〈0. 05). The activity of serum LDH increased and then decreased with the increasing time of ethanol exposure,the activity of serum LDH in the high-dose ethanol group was significantly lower than that in the control group four weeks after ethanol exposure( P〈0. 05). With the increasing time of ethanol exposure,the activity of serum ALP decreased in the control group,low- and middle-dose ethanol groups,but decreased and then increased in the high-dose ethanol group,the activity of serum ALP in the high-dose ethanol group was significantly higherthan that in the control group four weeks after ethanol exposure( P〈0. 05). With the increasing time of ethanol exposure,the activity of serum CHE increased in the control group,low- and middle-dose ethanol groups,but decreased in the high-dose ethanol group,the activity of serum CHE in the high-dose ethanol group was significantly lower than that in the control group in the second and fourth weeks after ethanol exposure( P〈0. 05).
