摘  要:目的探讨难治性癫痫细胞模型(Sombati癫痫细胞模型)中N-乙酰氨基葡萄糖转移酶Vb(GnT-Vb)和肌营养不良蛋白聚糖(dystroglycan)基因的表达变化及意义。方法取新生24 h内SD乳鼠,分离其双侧海马进行原代海马神经元培养。将培养至第14天的海马神经元分为对照组和模型组,模型组利用低镁细胞外液培养3 h制备Sombati癫痫细胞模型。通过实时定量聚合酶链式反应(real-time qPCR)方法测定对照组和模型组造模后第1天和第4天GnT-Vb和dystroglycan的基因表达水平。结果造模后第1天,模型组GnT-Vb基因表达水平低于对照组(P<0.05);而两组dystroglycan基因表达水平间无差异(P>0.05)。造模后第4天,模型组GnT-Vb、dystroglycan基因表达水平均高于对照组(P<0.05)。结论 GnT-Vb和dystroglycan基因在Sombati癫痫细胞模型中表达呈增高趋势,并呈现一定时间的动态变化;GnT-Vb和dystroglycan基因可能同时参与了难治性癫痫的形成过程。Objective To investigate the gene expression changes of GnT- Vb and dystroglycan in the Sombati cell culture model of refractory epilepsy.Methods The hippocampal neurons which were isolated from bilateral hippocamp of newborn SD rats within 24 hours of birth,were cultured in vitro.On the 14th day,the neurons were divided into control group and experimental group.The experimental group was cultured with low Mg2 +extracellular fluid for 3 hours to establish Sombati cell culture model.The real- time qPCR was used to detect the mRNA expression levels of GnT- Vb and dystroglycan in control group and experimental group on the first day and the fourth day.Results On the first day after model establishment,the mRNA expression level of GnT- Vb in experimental group was significantly lower than that in control group(P〈0.05).There was no significant difference in mRNA expression level of dystroglycan between two groups(P 〉0.05).On the fourth day after model establishment,mRNA expression levels of GnT- Vb and dystroglycan in experimental group were significantly higher than those in control group(P〈0.05).Conclusion The increasing trend of mRNA expression levels of GnT- Vb and dystroglycan can be observed in the Sombati cell culture model,and the expression levels change in a specific time period.The mRNA of GnT- Vb and dystrogly may participate in the formation process of refractory epilepsy.
