摘 要:目的检测钒氧化物、电离辐射、尘肺、黄磷四种接害工人外周血中EGFR和KRAS基因突变情况。方法分别对102名接触钒氧化物、164名接触电离辐射、88名接触黄磷工人以及78名尘肺患者进行问卷调查(性别、年龄、工种和接触时间),采集外周静脉血样品并提取DNA,采用聚合酶链式反应-限制性片段长度多态性分析(PCR-RFLP)检测EGFR基因外显子21中的L858R点突变(CTG→CGG,Leu→Arg)和KRAS基因外显子2上的G12C(GGT→TGT,Gly→Cys)、G12V(GGT→GTT,Gly→Val)和G12D(GGT→GAT,Gly→Asp)等点突变情况。结果四种接害工人外周血中均未检测到EGFR基因外显子21中L858R点突变和KRAS基因第12位密码子的点突变。结论钒氧化物、电离辐射、尘肺、黄磷四组接害工人外周血DNA均未发现EGFR基因外显子21中的L858R突变和KRAS基因外显子2中第12位密码子的点突变。Objective The aim of the study is to detect EGFR and KRAS mutations in peripheral blood of occupational workers exposed to 4 different hazards. Methods A questionnaire survey (including sex, age, occupation and contact time) was made in 102 workers exposed to vanadium oxides, 164 workers exposed to ionizing radiation, 88 workers exposed to yellow phosphorus and 78 pneumoconiosis patients. DNA was extracted from these workers' peripheral blood samples. PCR-RFLP was used to detect EGFR L858R mutation in exon 21 and KRAS mutations such as G12C (GGT→TGT, Gly→Cys), G12V (GGT→GTI→Gly→Val) and G12D (GGT→GAT, Gly→Asp) in exon 2. Results EGFR L858R mutation in exon 21 and KRAS point mutations in codon 12 had not been detected in peripheral blood of occupational workers exposed to 4 different hazards. Conclusion There is no EGFR L858R mutation in exon 21 and KRAS point mutations in codon 12 in peripheral blood of workers exposed to vanadium oxides, ionizing radiation, yellow phosphorus or pneumoconiosis patients.
