摘 要:目的观察慢性疼痛—激怒应激对大鼠血及结肠粘膜中血管活性肠肽(VIP)、神经肽Y(NPY)、胰高糖素(Glu)的影响。方法用夹子将大鼠后肢夹住,直立位挂起,并让大鼠之间出现扭打嘶咬;钳夹时间d1为30min,以后每天增加10mim,连续2wk;以不作任何处理的大鼠作正常对照。造模结束后血浆及结肠粘膜,以放免法测定血及结肠粘膜中VIP,NPY及Glu的含量。结果模型组大鼠血NPY含量(1045±608)ng·L^1,低于正常对照组(4627±1518)ng·L^1,P<0.01;模型组血VIP含量(93±27)ng·L^1,正常对照组(113±50)ng·L^1,P>0.05;模型组血Glu含量(82±28)ng·L^1,正常对照组(81±20)ng·L^1,P>0.05。模型组大鼠湿结肠粘膜VIP含量(9.78±4.98)pg·g^1,高于正常对照组(1.58±1.55)±pg·g^1,P<0.01;模型组湿结肠粘膜NPY含量(5.46±2.14)pg·g^1,高于正常对照组(0.84±0.39)pg·g^1,P<0.01;模型组湿结肠粘膜Glu含量(15.81±6.40)pg·g^1,正常对照组(18.76±6.62)pg·g^1,P>0.05。结论慢性应激可引起大鼠血NPY和结肠粘膜VIP和NPY的变化,这些变化可能在胃肠功能紊乱性疾病中起一定的作用。AIM To observe the effects of chronic pain-irritation stress on vasoactive intestinal peptide (VIP), neuropeptide Y (NPY) and glucagon (Glu) in rats blood and colonic mucosa. METHODS The rats were hung with clamp in their posterior limb, and made to wrestle and bite each other. Thirty minutes were taken in the first day, and ten minutes were added each day for two weeks. The group without clamping used as controls. After the models were made, the plasma and the colonic mucosa of all the rats were taken, and the contents of VIP, NPY and Glu were measured by RIA RESULTS The plasma NPY content ( 1045±608 ng·L^1 ) in the chronic irritation stress group was decreased more significantly than that in normal group (4627±1518 ng·L^1) (P<0.01); the plasma VIP content was (93±27 ng·L^1) in the model and (113±50 ng·L^1) in normal. There was no significant difference between the two groups (P>0.05); the plasma Glu content was (82±28 ng·L^1) in the model and (81±20 ng·L^1) in normal. The colonic mucosa VIP content ( 9.78±4.98 pg·g^1 ) in the model group was higher than that (1.58±1.55 pg·g^1) in the normal, P<0.01; the colonic mucosa NPY content (5.46±2.14 pg·g^1) in the model group was also higher than that (0.84±0.39 pg·g^1) in the normal, P<0.01; but the contents of Glu in the colonic mucosa had no significant difference between the model (15.81±6.40 pg·g^1) and the normal (18.76±6.62 pg·g^1). CONCLUSION NPY in the plasma, VIP and NPY in the colonic mucosa in the stressed rats have changed, and those changes might play some role in gastrointestinal functional disorders.
