灭幽汤治疗幽门螺杆菌相关性胃炎脾胃湿热证的机理研究

摘　　要：目的观察幽门螺杆菌相关性胃炎(HAG)脾胃湿热证小鼠模型TLR4、IL-6的表达,探寻灭幽汤治疗HAG脾胃湿热证的作用机理。方法将BABL/c小鼠分成空白组、模型组、灭幽汤组、胃三联组。空白组正常饲养;模型组使用复合因素(湿热环境、高糖高脂饲料、幽门螺杆菌感染)造成HAG脾胃湿热证小鼠模型;灭幽汤组和胃三联组分别使用清热祛湿的灭幽汤和胃三联药物对模型鼠进行治疗。采用免疫组化法检测胃黏膜组织TLR4蛋白的表达,实时荧光定量PCR检测胃黏膜TLR4 mRNA的表达,ELISA检测血清白介素(IL)-6。结果TLR4蛋白及mRNA,IL-6的表达,灭幽汤组、胃三联组均降低,与模型组比较差异显著(P<0.01);灭幽汤组与胃三联组比较无显著差异(P>0.05)。结论灭幽汤治疗幽门螺杆菌相关性胃炎脾胃湿热证的机理可能是干预TLR4、IL-6的表达从而降低胃黏膜组织的炎性程度。Objective： To observe the expression of TLR4 and IL-6 in rat model of helicobacter pylori associated gastritis with spleen-stomach damp-heat syndrome, and to explore mechanism of Mieyou Decoction treating helicobacter pylori associated gastritis with spleen-stomach dampheat syndrome. Methods： BABL/e rats were divided into control group,model group,Mieyou Decoction group,and Weisanlian group. The control group was given normal feeding while the model group was surrounded by composite factors （hot and humid environment,highsugar and high-fat diet,H, pylori infection） to make rat model of helicobacter pylori associated gastritis with spleen-stomach damp-heat syndrome. Mieyou Decoction group and Weisanlian group were given Mieyou Decoction and Weisanlian drugs respectively. Immunohistochemics and real time quantitative PCR were used to detect the TLR4 protein expression and TLR4 mRNA in gastric mucosa respectively,while ELISA was applied to detect IL-6 in serum. Results： The expression of TLR4 protein,and mRNA and IL-6 in serum in Mieyou Decoction group and Weisanlian group were lower compared with that in model group （P〈0.01 ）. Mieyou Decoction group has no statistical difference compared with Weisanlian group （P〉0.05）. Conclusion： The mechanism of Mieyou.Decoction treating helicobacter pylori associated gastritis with spleen-stomach damp-heat syndrome may prevent TLR4, IL-6 expression thereby reducing the degree of inflammation of gastric mucosa.
