摘 要:目的:观察不同肿瘤细胞系在仓鼠体内种植生长的差异,阐明不同胰腺癌细胞系的侵袭转移特性。方法:胰腺癌细胞系PC1.0、PC-1、MEK1/2行仓鼠胰腺原位种植、腹腔注射,建立不同的胰腺癌细胞侵袭动物模型。结果:高侵袭转移PC1.0组在肿瘤组织生长大小、腹水出现情况、肿瘤腹腔转移情况明显高于其他3组细胞;MEK1基因敲减组在胰腺种植、腹腔种植肿瘤组织生长比MEK2基因敲减组生长减小,腹膜及肝脏转移及周围脏器侵袭比MEK2基因敲减组增多。结论:PC1.0肿瘤生长旺盛且转移多;PC-1肿瘤生长一般且转移少;MEK1基因敲减肿瘤生长受抑制,但侵袭转移正常;MEK2基因敲减肿瘤生长不受抑制,但侵袭转移受抑制。Objective: To observe the difference growth of pancreatic cancer planted in hamsters. Clarfy the invade and metastasisis characteristic of pancreatic cancer. Methods: Established animal model with Pancreatic cancer cell system PC1.0; PC-1, MEK1 knowed down and MEK2 knowed down cell lines. Tumor cells was liant in animal pancreat and injected in abdomen. Results:Compare with PC1.0, the tumor grows, invasion, metastasis of MEK1 knowed down and MEK2 knowed down cell lines were restrain. MEK2 knowed down pancreatic tumor ceil displayed a weak- en invasion and metastasis ability than MEK1 knowed down pancreatic tumor cell, MEK1 knowed down pancreatic tumor cell displayed a weaken tumor grows than MEK2 knowed down pancreatic tumor cell. Conclusons: MEK1/2 gene inhibition inhibits the pancreatic cancer cell invasion and metastasis. MEK1/2 may serve as the candidate gene or potential target for the development of the new clinical pancreatic cancer drug.
