摘 要:目的比较3株来自我国不同地区的狂犬病病毒街毒株在细胞和乳鼠脑内的生长特性,为建立狂犬病病毒感染模型奠定基础。方法分别通过乳鼠脑内接种和细胞培养法进行连续传代,测定不同代次SXYQ、YN01和GDMMD57株狂犬病病毒街毒株的病毒滴度,计算半数细胞感染量(TCID50)。结果 SXYQ、YN01和GDMMD57株病毒乳鼠脑内接种传代至10代,病毒滴度随传代次数的增加,呈上升趋势,至第8代后,病毒滴度趋于稳定,分别为105.85、105.63和105.97 TCID50/g,不同毒株间病毒滴度存在差异。SXYQ、YN01和GDMMD57株病毒接种N2a细胞后,随着传代次数的增加,病毒对细胞的适应性不断增强,病毒滴度呈上升趋势,至第15代后,病毒滴度趋于稳定,不同毒株间病毒滴度存在差异;第20代SXYQ株病毒的滴度在感染后2.5 d达到峰值(106.80 TCID50/ml),YN01和GDMMD57株病毒滴度在感染后3 d达到峰值(均为107.30 TCID50/ml)。结论 3株病毒在细胞和乳鼠脑内经连续传代后均能达到较高滴度,可用于狂犬病病毒感染模型的建立及新型疫苗的研究。Objective To compare the growth characteristics of three street rabies virus strains(SXYQ,YN01 and GDMMD57)isolated in China in cells and suckling mouse brain,and lay a foundation of the establishment of rabies virus infection model. Methods The three strains were subcultured in suckling mouse brain and in N2a cells respectively,of which the various passages were determined for infectious titers,and calculated for TCID50. Results The titers of SXYQ, YN01 and GDMMD57 strains subcultured in suckling mouse brain showed increasing tendencies with the increasing passage within passage 10,and were stable as 10^5. 85,10^5. 63 and 10^5. 97 TCID50/ g respectively after passage 8,which showed significant difference. However,the adaptabilities of SYXYQ,YN01,GDMMD57 strains in N2a cells increased with the increasing passage,while the titers showed increasing tendencies and were stable after passage 15,which showed significant difference. The titer of SXYQ strain of passage 20 reached a peak value of 106. 80TCID50/ ml 2. 5 d, while those of YN01 and GDMMD57(both 10^7. 30 TCID50/ ml) strains 3 d after infection. Conclusion All the three rabies virus strains reached high titers after subculture in both suckling mouse brain and cells,which might be used for the establishment of rabies virus infection model and development of novel rabies vaccines.
