摘 要:目的探讨HMGA1通过Wnt/β-Catenin通路在胃肿瘤形成中的作用。方法应用小干扰RNA介导的基因沉默、细胞增殖分析、PCR等技术完成实验。结果 HMGA1的特异敲除明显减少细胞生长。β-Catenin或下游c-myc的损失减少HMGA1表达,而Wnt3a处理增加HMGA1和c-myc的转录。结论这些数据表明,HMGA1参与胃癌细胞形成和增殖通过Wnt/β-Catenin通路。Objective To investigate the role of HMGA1 by Wnt/β-Catenin pathway in gastric tumor formation. Methods Adenovirus transfer, cell proliferation assay, and RT-PCR were used to perform the experiment. Resuits Specific knockdown of HMGA1 resulted in marked reduction of cell growth. The loss of β-catenin or its downstream c-myc decreased HMGA1 expression, whereas Wnt3a treatment increased HMGA1 and c-myc transcripts. Furthermore, Wnt/β-induced expression of HMGA1 was inhibited by c-myc knockdown, suggesting that HMGA1 is a downstream target of the Wnt/β-catenin pathway. Enhanced expression of HMGA1 coexisted with the nuclear accumulation of a-catenin in about 30% of gastric cancer tissues. Conclusion These data suggest that HMGA1 is involved in the proliferation and gastric tumor formation via the Wnt/β-catenin pathway.
