摘 要:目的 研究alpha-黑素细胞刺激素(α-MSH)及其新型类似物(STY39)对内毒素血症小鼠产生组织因子(TF)的影响.方法 BALB/c小鼠经腹腔注射LPS (25 μg/kg)和D-Gal(100 mg/kg)建立内毒素血症模型,分别在LPS刺激后1、2、3h经腹腔注射α-MSH(2.5 mg/kg)或STY39(2.5 mg/kg),于注射后6h采血并颈椎脱臼处死小鼠,取肝、肺、肾等组织.ELISA检测小鼠血清TF浓度;RT-PCR检测小鼠肝、肺、肾组织中TF mRNA表达,从蛋白、基因两个水平检测与评价α-MSH和STY39对TT表达的影响.结果 内毒素血症小鼠血清TT浓度明显升高,肝、肺、肾组织中TF mRNA的表达水平增高,在不同时间点给予α-MSH或STY39,均能降低小鼠血清TF浓度,抑制肝、肺、肾组织中TF mRNA的表达,且在LPS刺激之后1h给药效果较理想.结论 α-MSH和STY39均能抑制内毒素血症小鼠TF的产生,且STY39优于α-MSH.Objective To evaluate the effect of alpha-melanocyte stimulating hormone (α-MSH) and its novel analogue STY39 on the production of tissue factor (TF) in the mice with endotoxemia.Methods Endotoxemia was modeled by LPS (25 μg/kg) and D-Gal (100 mg/kg)injection intraperitoneally of 6-week-old female BALB/c mice.The serum TF levels were determined by ELISA,and the expression of TF mRNA in different tissues was tested by RT-PCR after treatment with 2.5 mg/kg α-MSH or STY39 at 1,2,3 h following LPS injection.Results LPS stimulation for 6 h resulted in an increase of serum TF level and high expression of TF mRNA in the different tissues from endotoxemia mice,especially in the livers,lungs and kidneys.The serum TF levels and TF mRNA expression in lungs,livers and kidneys were reduced markedly in α-MSH or STY39 treated endotoxemia mice,especially the administration of α-MSH or STY39 in 1 h after LPS injection.Conclusion α-MSH and STY39 suppress the production of TF in the endotoxemia mice,and the effect of STY39 is superior to α-MSH.
