摘 要:目的探讨红细胞CD35表达数量与红细胞免疫黏附肿瘤细胞活性相关性,研究恶性肿瘤患者红细胞CD35表达数量及免疫黏附功能的变化规律。方法选择原发性肺癌40例,胃癌32例,结肠癌36例及40例健康人血标本,用流式细胞术(FCM)检测红细胞CD35几何平均荧光强度(CMFI);EAC花环试验检测红细胞免疫黏附肿瘤细胞花环率。结果肺癌、胃癌、结肠癌组较正常对照组红细胞黏附肝癌细胞BEL-7402花环率下降,t值分别为3.069、3.429、3.056,P值分别为0.004、0.002、0.004,差异有统计学意义(P<0.01);肺癌、胃癌、结肠癌组间花环率比较,F=0.844,P=0.433,差异无统计学意义(P>0.05)。肺癌、胃癌、结肠癌组较正常对照组红细胞CD35 GMFI降低,t值分别为3.392、2.767、3.635,P值分别为0.002、0.009、0.001,差异有统计学意义(P<0.01);肺癌、胃癌、结肠癌组间红细胞CD35 GMFI比较,F=0.717,P=0.491,差异无统计学意义(P>0.05)。肺癌、胃癌、结肠癌、正常对照组红细胞黏附肿瘤花环率与其红细胞CD35 GMFI呈正相关,相关系数分别为0.628、0.859、0.636、0.887,P值分别为0.000、0.000、0.000、0.000,有统计学意义(P<0.01)。结论肺癌、胃癌、结肠癌组红细胞黏附功能下降;肺癌、胃癌、结肠癌组红细胞CD35表达降低;肺癌、胃癌、结肠癌、正常对照组红细胞黏附肿瘤花环率与其细胞CD35 GMFI呈正相关;流式细胞术(FCM)检测红细胞CD35的表达是一种准确、快速、高效的肿瘤患者红细胞免疫功能检测方法。Objective To investigate the correlation between CD35 expression of erythrocytes and erythrocyte immune adherence activity to tumor cells.Methods The geometric average fluorescence intensity (CMFI) of CD35 of erythrocytes and the erythrocyte immune adherence rosette rate were examined separately with flow cytometry (FCM) and EAC rosette experiments in peripheral blood of 40 patients of lung cancer,32 patients of gastric cancer,36 patients of colon cancer and 40 healthy human.Results The erythrocyte adhesion rosette rate which adhered to liver cancer cells (BEL-7402) in the groups of lung cancer,stomach cancer,colon cancer were lower than that in the control group (The t values were 3.069,3.429,3.056 respectively;P values were 0.004,0.002,0.004 respectively;P 〈0.01).The erTthrocyte adhesion rosette rate was not significantly different with each other in the groups of lung cancer,stomach cancer,colon cancer(F value was 0.844; P value was 0.433; P 〉0.05).The CMFI of CD35 in groups of lung cancer,stomach cancer and colon cancer was lower than that in the control group(The t values were 3.392,2.767,3.635respectively;P values were 0.002,0.009,0.001 respectively;P 〈0.01).The CMFI of CD35 in cancer groups was not significantly different with each other(F value was 0.717;P value was 0.491 ; P 〉 0.05).The rosette rate of erythrocytes adhesion was positively related to the GMFI of erythrocytes (The r values were 0.628,0.859,0.636,0.887 respectively ;P values were 0.000,0.000,0.000,0.000respectively ;P 〈 0.01).Conclusions The erythrocytes adhesion function in the group of lung cancer,stomach cancer and colon cancer decline,simultaneously,the expression of CD35 of erythrocytes decline too.What is more,the rosette rate of erythrocytes adhesion is positively related to the GMFI of erythrocytes.It is an accurate,fast,and efficient method to evalulate the erythrocyte immune function by detecting the expression of CD35 of erythrocytes with FCM.
