摘  要:目的 探讨密蒙花总黄酮对去势雄鼠干眼症动物模型泪腺组织超微结构的变化.方法 150只健康1月龄Wistar雄性大鼠随机分为5组:正常组(A)、假手术组(B)、模型对照组(C)、雄激素治疗组(D)、密蒙花总黄酮治疗组(E),每组30只.各组分别于用药1,3,5月后随机取10只,行Schirmer I试验、泪膜破裂时间及角膜荧光素染色检查,并取双眼泪腺进行透射电镜观察.结果 C组3月起角膜荧光素染色阳性,且随实验时间延长而加重(P<0.05);D、E组于去势3,5月后,角膜荧光素染色点较C组减少,SIT、BUT明显高于C组(P<0.05);D、E两组间5月后,差异有显著性(P<0.05).D、E两组间泪腺组织 超微结构改变较C组相比明显减轻.结论密蒙花总黄酮能减轻去势雄鼠逐渐加重的泪腺分泌功能损害和角膜上皮缺失,对干眼症模型泪腺组织有一定的保护作用.Objective To assess the effects of buddleja officinalis flavones on microstructure changes of lacrimal gland in castrated rats with dry eye. Methods One hundred and fifty healthy Wistar rats were divided randomly into 5 groups each with 30 rats; the normal group(A) ; the false operation group(B) ; the model group (C); the therapy group treated with testosterone (D); the therapy group with buddleja officinalis flavones(E). Ten experimental animals in each group were randomly chosen after 1 month,3 and 5months medication respectively, which were performed Schirmer I test, tear film break-up time and corneal fluorescein staining. And then the animals were killed to observe the cornea and lacrimal glands with transmission electron microscopy. Results After 3 months the cornea [luoreseein staining of group C were positive and aggravated as times(P was less than 5 percent); after 3,Smonth the cornea fluorescein staining of group D,E were less than that of group C. While SIT and BUT of group D,E were much more than that of group C (P was less than 5 percent). After 5months there was significant difference between group D and E. (P was less than 5 percent). The microstructure changes of lacrimal gland of group D and E were less than those of group C. Conclusion Buddleja officinalis flavonoids can reduce the gradually increasing damage of lacrimal gland and loss of corneal epithelial of the castrated male rats and protect dry eye model of the lacrimal gland.
