摘  要:目的:研究FGF-23(R176Q)过表达对成年小鼠磨牙龋损的影响及机制。方法:使用CT扫描、血清学检查、HE染色、免疫组化染色和real time RT-PCR等方法检测分析FGF-23(R176Q)转基因小鼠(TG组,n=20)和野生型小鼠(WT组,n=20)血清钙磷浓度变化和下颌第一磨牙矿化的差异。结果:WT组小鼠血清钙、磷和1,25(OH)2D3浓度均明显高于TG组(P<0.05),PTH的浓度低于TG组(P<0.05);TG组下颌第一磨牙龋损率高于WT组(P<0.05),前期牙本质厚度高于WT组(P<0.05),而继发性牙本质面积低于WT组(P<0.05);TG组的Biglycan阳性百分比高于WT组(P<0.05),而DSP阳性百分比低于WT组(P<0.05);WT组的ALP和Ⅰ型胶原mRNA表达水平均明显高于TG组(P<0.05)。结论:FGF-23(R176Q)过表达能够抑制牙本质基质形成,减少前期牙本质形成和矿化,减少继发性牙本质形成,进而导致了龋损增加。Objective: To study the effects of FGF-23 (R176Q) overexpression on the caries development of adult mouse molar. Methods: CT scanning, serological examination, HE staining, immunohistochemical staining and real time RT-PCR analysis were used to compared the serum calcium, phosphorus concentrations and mandibular first molar mineralization of FGF-23 (R176Q) transgenic mice (TG group, n = 20 ) and wild type mice( WT group, n = 20 ). Results: Caries prevalence in TG group was higher than that in WT group ( P 〈 0.05 ). Serum calcium, phosphorus and 1,25 ( OH ) 2 D3 concentrations in WT group were higher and PTH was lower than those in TG group( P 〈 0.05 ) ; the predentin thickness of TG group was higher while the secondary dentin area was smaller than those of WT group (P 〈 0.05 ) ; the positive percentage of Biglycan of TG group was higher while the DSP positive percentage was lower than those of WT group(P 〈 0.05 ) ; the ALP and collagen I mRNA levels in the WT group were higher than those in TG group(P 〈 0.05). Conclusion: FGF-23 (R176Q) overexpression can inhibit the formation of dentin matrix, reduce predentin mineralization, reduce the formation of secondary dentin and increase dental caries.
