摘 要:目的:验证鱼腥草素钠体外对白色念珠菌生物膜的抑菌效果,并初步研究其抑菌作用是否为通过影响或干扰毒力因子磷脂酶的表达而实现的。方法:建立白色念珠菌生物膜的体外模型,MTT法计算鱼腥草素钠对白色念珠菌生物膜的抑菌率,然后用RT-PCR方法检测白色念珠菌毒力因子PLB1、PLB2和PLC1受鱼腥草素钠作用后的表达水平的变化。结果:鱼腥草素钠对白色念珠菌生物膜的抑制作用随药物浓度增加而增加。RT-PCR结果表明,毒力因子PLB1、PLB2的表达水平随着药物浓度的梯度增加而降低,在浓度为240mg/L时mRNA的表达水平受到完全抑制,而药物浓度变化对PLC1的表达影响较小。结论:鱼腥草素钠体外对白色念珠菌生物膜有一定的抑制作用,并对本实验所选毒力因子PLB1、PLB2的表达过程明显产生了抑制作用。Objective: To confirm the antibacterial effect of sodium houttuyfonate on candida albieans biofilms and learn whether the inhibition is achieved by affecting or interfering the expression of phospholipase genes. Methods: We established the biofilms model of the candida albicans in vitro, calculated the inhibition rate of sodium houttuyfonate to the candida albicans biofilms by MTT. Then the expressions of virulence genes PLB1,PLB2 and PLC1 transcription levels were determined using RT--PCR method. Results: Inhibitory effect of sodium houttuyfonate to the candida albicans biofilms increased with the drug concentration. RT--PCR results showed that the expression levels of virulence factors PLB1 and PLB2 decreased with the concentration gradient increased, and the mRNA ex- pression levels were completely inhibited at a concentration of 240vg/mL. Drug concentration change on the expres- sion of PLC1 had less effect. Conclusion: The expression of the selected pathogenic genes PLB1 and PLB2 as well as the growth of candida albicans biofilms were inhibited by sodium houttuyfonate.
