摘  要:目的研究钙结合蛋白S100A11在小鼠胚胎种植早期子宫内膜中的表达及该蛋白在子宫内膜容受性中的作用。方法采用免疫组织化学法、Real-time PCR和Western blot法分别检测钙结合蛋白S100A11在妊娠早期子宫的细胞定位、mRNA和蛋白表达情况。结果免疫组化结果显示S100A11蛋白主要分布于子宫内膜腺上皮和腔上皮;Real-time PCR结果表明S100A11mRNA在围着床期均有表达,其中妊娠第4天表达量最高;Western Blot表明S100A11蛋白在围着床期子宫中均有表达,并在妊娠第4天、第5天明显升高。结论钙结合蛋白S100A11调节了胚胎在子宫内膜上的黏附与侵入,参与了子宫内膜的容受性及免疫耐受。Objective It is to research the expression of calcium-binding protein S100A11 in mouse endometrium in early embryo plantation stage and the function of this protein in endometrial receptivity. Methods Real-time PCR was used to detect the concentration of S100A11 mRNA, immunohistochemical and western blot method was used to detect the concentration of S100A11 protein expression in the uterine tissue in peri-implantation. Results The immunohistochemical results showed that S100A11 was mainly located in luminal and glandular epithelium of endometrium. Real-time PCR results indicated that the expression of S100A11 mRNA could be found during peri-implantation, in which the expression was the highest on the fourth day of pregnancy. Western blot results prompted that the expression S100A11 could be found in the implantation window phase of endometrium, which was much higher on the fourth day and fifth day of pregnancy than that of other days. Conclusion The specific expression of S100A11 gene in endometrium regulates the adhesion and invasion of embryo, and it can be participated in the formation of endometrial capacity and immune tolerance.
