摘 要:目的探讨miR-10b、miR-18b及miR-130b在肺癌中的表达特征及其在肺癌发生发展过程中的作用。方法收集47例肺癌住院患者的癌组织和癌旁组织,利用Trizol提取总RNA,实时荧光定量PCR检测基因相对表达量。结果肺癌组织与癌旁组织中miR-10b(t=2.83,P<0.05)、miR-18b(t=2.88,P<0.05)及miR-130b(t=2.46,P<0.05)表达量均差异存在显著性。比较鳞癌、腺癌和癌旁组织中miR-10b(F=7.16,P<0.05)、miR-18b(F=4.11,P<0.05)及miR-130b(F=3.43,P<0.05)的表达量也有显著性。癌组织中miR-10b与pri-miR-10b表达量存在负相关关系(r=-0.50,P<0.05)。癌组织与临床指标之间的关系中miR-18b、miR-130b与KI67呈正相关(P<0.05),pri-miR-10b与TOPOⅡ、pri-miR-130b与MRP呈负相关(P<0.05)。结论 miR-10b、miR-18b及miR-130b在癌组织中表达量上调,可能具有促进肺癌发生发展的作用。【Objective】To analyze the expression characteristics of miR-10b, miR-18b, miR-130b and explore their roles in the occurrence and development of lung cancer. 【Methods】47 cases of hospitalized patients were chose with lung cancer tissues and adjacent tissues, total RNA was extracted with Trizol reagent, the gene relative expression level was detected by fluorescence real-time quantification PCR.【Results】The expression of miR-10b(t =2.83, P &lt;0.05), miR-18b(t =2.88, P &lt;0.05) and miR-130b(t =2.46, P &lt;0.05) had statistical significance between lung cancer tissues and adjacent tissues, moreover, miR-10b(F =7.16, P &lt;0.05), miR-18b(F =4.11, P &lt;0.05) and miR-130b(F =3.43, P &lt;0.05) were also statistically significant among the squamous carcinoma tissues, adenocarcinoma tissues and adjacent tissues. In lung cancer tissues, the correlation analysis showed a negative correlation in miR-10b and pri-miR-10b(r =-0.50, P &lt;0.05), the relationship also existed in pri-miR-10b and TOPOⅡ, primiR-130b and MRP(P &lt;0.05), while the miR-18b, miR-130b and KI67 had a positive correlation, respectively(P &lt; 0.05). 【Conclusion】The expression of miR-10b, miR-18b and miR-130b were up-regulated in cancer tissues,they might play a role promoting the occurrence and development of lung cancer.
