Siah2调控脯氨酰羟化酶3表达在非小细胞肺癌中的临床意义

摘  要:目的观察Siah2(seven in absentia2)及脯氨酰羟化酶3(prolyl hydroxylase 3,PHD3)在人非小细胞肺癌及癌旁组织中的表达,并探讨上述两者差异表达在非小细胞肺癌发病过程中的临床意义。方法应用RT-PCR检测非小细胞肺癌患者89例肿瘤及癌旁正常肺组织标本Siah2、PHD3 mRNA表达情况,Western blot检测上述指标的蛋白质表达水平,并分析其与肺癌临床特征的关系。结果非小细胞肺癌患者肿瘤组织中Siah2蛋白及mRNA表达量均明显高于癌旁组织(P<0.05),PHD3蛋白的表达在肿瘤组织中低于癌旁组织(P<0.05),PHD3 mRNA在肿瘤组织中的表达高于癌旁组织,Siah2蛋白与PHD3蛋白表达呈负相关(r=-0.735,P<0.05),Siah2蛋白与Siah2 mRNA表达呈正相关(r=0.821,P<0.05)。结论 Siah2可能通过调控PHD3表达水平在非小细胞肺癌发病中发挥作用。Objective To investigate the expression of Siah2(seven in absentia2) and prolyl hydroxylase 3(PHD3) in non-small cell lung cancer ( NSCLC) ,and to elucidate their clinical significance .Methods The mRNA expression levels of Siah2 and PHD3 were detected by RT-PCR in 89 cases of NSCLC and adjacent normal lung tissues , and their protein expression levels were detected by Western Blot .Results The expression levels of Siah 2 mRNA and protein in tumor tissues were significantly higher than those in adjacent tissues ( P 〈0.05).The expression levels of PHD3 mRNA in tumor tissues were obviously higher than those in adjacent tissues , however , the expression levels of PHD 3 protein in tumor tissues were significantly lower than those in adjacent tissues ( P 〈0.05).The expression of Siah2 protein was negatively correlated to that of PHD3 protein( r =-0.735, P 〈0.05),however,the expression of Siah2 protein was positively related with that of Siah 2 mRNA( r =0.821, P 〈0.05).Conclusion Siah2 may play a role in pathogenesis of NSCLC by regulating PHD 3 expression levels.
