摘  要:目的:探讨肝细胞生长因子(HGF)对体外缺血/再灌注导致的星形胶质细胞氧化应激的影响.方法:取体外培养的大鼠大脑皮层星形胶质细胞,建立缺血/再灌注损伤细胞模型.用HGF预处理星形胶质细胞后,进行缺血/再灌注.测定细胞活力、活性氧簇(ROS)水平、丙二醛(MDA)含量、超氧化物歧化酶(SOD)活性及谷胱甘肽(GSH)含量.RT-PCR检测细胞Nrf2 mRNA的表达,Western blotting检测胞浆和胞核Nrf2蛋白的表达.结果:星形胶质细胞缺血/再灌注后,细胞活力降低,ROS水平及MDA含量升高,SOD活性及GSH含量降低.Nrf2 mRNA及胞浆、胞核Nrf2蛋白水平均下调.HGF能明显升高细胞活力,降低ROS水平及MDA含量,升高SOD活性及GSH含量,上调Nrf2 mRNA及胞浆、胞核Nrf2蛋白的表达.结论:HGF能减轻体外缺血/再灌注导致的星形胶质细胞氧化应激损伤,可能与HGF调节Nrf2表达及核转位水平、调控氧化/抗氧化系统的平衡有关.Aim:To explore the effect of hepatocyte growth factor (HGF)on the oxidative stress of as-trocytes resulting from ischemia/reperfusion in vitro.Methods:Cultured cerebral cortical astrocytes were prepared from rats,and the ischemia/reperfusion cell model in vitro was established.After pretreatment with HGF,the cultures were exposed to ischemia /reperfusion.Then cell viability,reactive oxygen spe-cies (ROS ) level and malondialdehyde (MDA ) content were measured.The superoxide dismutase (SOD)activity and glutathione (GSH)content were evaluated.The expression of Nrf2 mRNA was as-sessed by RT-PCR.The protein expression of Nrf2 in cytoplasm and nucleus were detected by Western blotting analysis.Results:Ischemia/reperfusion decreased cell viability,increased ROS level and MDA content,decreased SOD activity and GSH content.The expression of Nrf2 mRNA,Nrf2 protein in cyto-plasm and nucleus were decreased.In the presence of HGF,cell viability was increased,ROS level and MDA content were decreased,SOD activity and GSH content were increased.In addition,HGF upregu-lated the expression of Nrf2 mRNA,Nrf2 protein in cytoplasm and nucleus.Conclusion:HGF can allevi-ate oxidative stress injury of astrocytes exposed to ischemia /reperfusion in vitro,which may be related to the regulation of Nrf2 expression,Nrf2 nuclear translocation,and the balance of oxidation/antioxidant system.
