摘  要:目的研究3,4’,5-三羟基芪-3-β-单-D-葡萄糖苷(3,4’,5,-trihydroxystibene-3-β-mono-D-glucoside,THMG)对大肠癌细胞粘附性、浸润力及蛋白激酶C活性的影响;探讨THMG抗转移作用的信号转导机制。方法利用Boyden小室法等细胞生物学技术检测4种大肠癌细胞(HR8348,Hce-8693,HT29,LoVo)在0.4,0.8,1.6和3.2mmol·L^(-1)等浓度THMG作用下的粘附率、浸润力。[γ-^(32)P]ATP掺入法测定蛋白激酶C活性。结果在3.2mmol·L^(-1)浓度时THMG对HR-8348,Hce-8693,HT-29和LoVo粘附抑制率分别为75.7%,69.4%,67.6%和78.8%,浸润抑制率分别为82.7%,85.7%,89.7%和92.7%,且随着浓度加大,粘附和浸润抑制率提高。3.2 mmol·L^(-1)THMG对HR-8348,Hce-8693,HT-29和LoVo的蛋白激酶C活性抑制率分别是35.1%,36.8%,41.5%,49.6%,呈剂量依赖性关系,与对照组比较具有统计学显著性(P<0.001)。粘附性及浸润力抑制程度与蛋白激酶C活性的抑制程度呈显著性相关(P<0.05)。结论 THMG可能通过抑制蛋白激酶C活性对信号转导系统具有调变作用,THMG对信号转导系统的影响可能是抗粘附抗侵袭效应的重要机制之一。AIM To investigate anti-adhesion and anti-invasion effects of 3,4, 5-trihydroxystibene-3-β-mono-D-glucoside (THMG) and protein kinase C (PKC) activity on the colorectal carcinoma cell lines in vitro. METHODS Four cell lines (HR8348, Hca8693, HT29 and LoVe) from human colorectal carcinoma were treated with 0.4, 0.8, 1.6 and 3.2 mmol·L^(-1) of THMG. The modified human amniotic basement membrane model and the Boyden chamber model were used for adhesion tests and quantitative invasion assay respectively, and incorporation of phosphate from [γ-^(32)P] ATP to histone for the measurement of the PKC activity. RESULTS At the experimental concentration, HMG had significant effects in terms of adhesion and invasion on four colorectal carcinoma cell lines (HR8348, Hca8693, HT29 and LoVe), with an adhesion inhibition rate of 75.7%, 69.4%, 67.6% and 78.8%, at the concentration of 3.2mmol·L^(-1), and with an invasion inhibition rate in Matrigal invasion assay of 82.7%, 85.7%, 89.7% and 92.7%, for those four cell lines. The adhesion inhibition rate and invasion inhibition rates were raised according to the concentration of THMG. PKC activity inhibition rates by THMG at the concentration of 3.2 mmol·L^(-1) were 35.1%, 36.8%, 41.5%, and 49.6%, respectively. Adhesion inhibition and invasion inhibition were significantly correlated with the PKC activity inhibition (P<0.05). CONCLUSION THMG, an effective component from Chinese herb (Polygormm Cuspidatum Sieb. et Zucc ), have anti-adhesion and anti-invasion function by inhibiting PKC activity on the colorectal carcinoma cell lines.
