摘 要:目的采用反相高效液相色谱法,建立杜仲叶的HPLC指纹图谱,比较杜仲雄株叶和雌株叶中几种次生代谢产物的含量。方法采用Ameritech C18(250 mm×4.6 mm,5μm)色谱柱,甲醇-0.05%磷酸溶液梯度洗脱;体积流量1.0 ml/min;检测波长238 nm,柱温25℃。结果对同一日期采集的杜仲叶指纹图谱进行评价,生成对照图谱,确定了19个共有峰。杜仲雄株叶中绿原酸含量略高于杜仲雌株叶,杜仲雄株叶中京尼平苷酸含量和京尼平苷含量略低于杜仲雌株叶,但差异不明显。结论性别对杜仲叶中几种次生代谢产物的含量无明显影响。Objective Use reversed - phase of HPLC to establish HPLC fingerprint of Eucommia ulmoides leaf and compare the content of several secondary metabolites in Eucommia ulmoides male and female leaves. Methods RP - HPLC method was used for the determination. The separation was performed on an Ameritech C18 column (250 mm×4.6 mm,5 μm), the mobile phase was methanol with 0.05% phosphoric acid in the gradient elution mode. The flow rate was 1.0mL/min. The detective wavelength was set at 238nm. The temperature of column was maintained at 259C. Results The fingerprint of samples from same picking time and same place were used to get the reference chromatogram composed of nineteen common peaks. The chlorogenic acid content in Eucommia ulmoides male leaf was slightly higher than Ettcommia ulmoides female leaves ; the geniposidic acid and geniposide contents were slightly lower than Eucommia female leaves, but the difference is not obvious. Conclusion the gender has no significant effect on the content of several secondary metabolites in Eucommia ulmoides leaves.
