摘 要:研究利用尿酸的特异性反应 ,采用差示光谱采样 ,用褶合曲线分析法进行数据处理测定血清尿酸含量。结果表明 ,尿酸浓度在 0~ 2 0 0μg· ml-1范围内 ,线性方程 Qij=0 .2 1 0 4 5+0 .0 0 1 864C,平均回收率 99.86% ,RSD为 0 .2 9% ;样品测定结果与尿酸酶法无显著性差异。该法可消除背景干扰且简便、实用、结果准确。A new, convenient and accurate method to determine uric acid in serum is reported. In an alkaline medium, uric acid in the sample is oxidized by potassium ferricyanide. The oxidized sample solution is taken as the reference solution, and the non oxidized sample solution (i.e. the acidic potassium ferricyanide solution is added) is taken as the measuring solution. The differential absorption spectrum is obtained by scanning in the wavelength range of 280~350nm at an interval of 1nm using the Cary 100 UV VIS spectrophotometer. By using the convolution curve method and with the aid of computer, the data treatment was carried out satisfactorily, and the background interference is also eliminated. The linear regression equation for the concentration range of 0~200μg·ml -1 of uric acid is obtained: Q ij =0 21045+0.001864 C . The average recovery is 99.86% and the RSD is 0.29%. The results obtained by this new method were compared with those by the conventional uricase method, and no significant differences were found between them. This method is proved to be simple, convenient, practical and accurate.
