摘  要:目的 探讨脑梗死后胆碱能神经系统的变化特点。方法 利用血管内栓线法制备大鼠右侧大脑中动脉栓塞(MCAO)模型,于MCAO后0.5h、1h、2h、3h、6h分别进行光镜乙酰胆碱酯酶(AChE)染色及病理染色,并进行电镜AChE活性的观察。结果 光镜下MCAO 后1h AChE活性开始下降,以第IV细胞层染色优势消失为特点,自2h随梗死时间延长,酶活性进一步下降。电镜下正常组织AChE酶颗粒主要分布在突触前后膜及突起中,半暗区AChE酶颗粒数量及密度降低,梗死区酶颗粒几乎消失。结论 急性脑梗死后AChE活性变化出现在形态学改变之前,且与细胞的损伤密切相关,脑梗死后较早即发生了胆碱能神经系统的异常。Objective To study the dynamic changes of cholinergic system after cerebral infarction. Methods A rat model of right middle cerebral artery occlusion(MCAO) was made by insertion of an intraluminal suture. The enzyme histochemistry study of Acetylcholine esterase (AChE) was done at 0.5h、1h、2h、3h、6h respectively after MCAO under light microscope. The activity changes of AChE under electron microscope were also done. Results Under light microscope the activity of AChE began to change at 1h after MCAO, showing th e weakening of staining priority in the fourth cell layer, and continued to decrease with MCAO time extending from 2h after MCAO. Under electron microscope, the AChE enzyme particles were located in the processes、the anterior and posterior membranes of the synapses which decreased both in quantity and density in the ischemic penumbra after MCAO, and almost disappeared in the infarcted core. Conclusion The activit y of AChE changes before morphologic changes appeared after MCAO.The cholinergic system become abnormality in the early stage after cerebral infarction.
