摘 要:目的研究miRNAs在乳腺癌患者血清中的异常表达及其作为潜在诊断标志物的价值。方法将乳腺癌患者纳入观察组,并分为TNMⅠ、TNMⅡ、TNMⅢ、TNMⅣ期,选择乳腺活检阴性患者作为对照组,采集血清检测MicroRNAS(简称miRNAS)含量。结果观察组患者miR-339、miR-21、miR-155、miR-103的含量高于对照组的,呈现出TNMⅠ<TNMⅡ<TNMⅢ<TNMⅣ期,且与TNM分期呈正相关;miR-145、miR-96、miR-129的含量低于对照组,呈现出TNMⅠ>TNMⅡ>TNMⅢ>TNMⅣ期,且与TNM分期呈负相关。结论乳腺癌组织中miRNA的表达量存在异常,TNM分期越差,miR-339、miR-21、miR-155、miR-103的含量越高,miR-45、miR-96、miR-129的含量越低,可作为乳腺癌的诊断标志物。Objective To study the abnormal expression of miRNAs in the serum of breast carcinoma patients and its potential value as diagnostic biomarker. Methods Breast carcinoma patients were enrolled as observation group and divided into TNMⅠ,TNMⅡ,TNMⅢ,TNMⅣ, and breast biopsy negative patients were collected as control group. Then serum were collect- ed from these patients to detect the miRNA level. Results The levels of miR - 339, miR - 21, miR - 155, miR - 103 in ob- servation group were higher than those in control group, and showed the order of TNMⅠ〈TNMⅡ〈TNMⅢ〈TNMⅣ, indi- cating the positive correlation with TNM stage. While the levels of miR - 145, miR - 96, miR - 129 in observation group were lower than control group, showed the order of TNMⅠ〉TNMⅡ〉TNMⅢ〉TNMⅣ, indicating the negative correlation with TNM stage. Conclusion miRNAs were abnormally expressed in breast carcinoma, as the TNM stage gets worse, the expres- sion levels of miR - 339, miR - 21, miR - 155, miR - 103 get higher and those of miR - 45, miR - 96, miR - 129 get lower, so miRNAs can be used as diagnostic biomarker of breast carcinoma.
