摘 要:目的对肝细胞癌(hepato celluar carcinoma,HCC)及其癌旁异型增生肝细胞Ki-67与α-甲酰基-辅酶A消旋酶(alpha-methylacyl-coenzyme A racemase,AMACR)和RAS表达进行定位分析,探讨AMACR的表达在HCC发生中的意义。方法 90例人HCC、90例癌旁肝组织制作组织芯片,以20例正常肝组织对照。用免疫荧光双标法,通过激光共聚焦显微镜,分析AMACR在肝细胞癌和癌旁异型增生肝细胞的表达与Ki-67、癌基因蛋白RAS表达的原位定位关系。结果 HCC组织中AMACR的表达显著高于癌旁肝组织,而且在HCC组织中AMACR的表达多呈弥漫性分布;HCC组织中显示Ki-67标记指数比癌旁组织高。结论 AMACR可能参与RAS信号在肝细胞异型增生和HCC早期发生的信号机制。Objective To investigate the localization of Ki-67,alpha-methylacyl-coenzyme A racemase( AMACR) and RAS-positive cells in hepatocellular carcinoma( HCC) and pericancerous dysplasia by double-labelled immunofluorescence staining. Methods Samples w ere collected from 90 cases of hepatocellular carcinoma and corresponding pericancerous liver tissues,and 20 cases of normal liver tissue from non-neoplastic disease. The tissue specimens w ere stained by double-labelled immunofluorescence for Ki-67,RAS and AMACR on self-made microarray,and observed under laser confocal microscopy. The relationship of AMACR expression and Ki-67 / RAS in HCC and pericancerous tissue w as analyzed. Results AMACR w as diffusedly distributed in most of HCC cells and it w as higher expressed in HCC than in pericancerous tissue. The labelling index of Ki-67 w as higher in HCC than in pericanerous tissue. Conclusion AMACR may participate in RAS signal pathw ay w hich involved in hepatocyte dysplasia and hepatocellular carcinogenesis.
