摘　　要：【目的】阐明BMP2是否参与了Decorin诱导的血管平滑肌细胞(VSMC)钙化。【方法】本研究采用人血管平滑肌细胞钙化体外模型,用腺病毒载体过表达Decorin,检测骨相关蛋白BMP2、Msx2、Osterix的表达和细胞钙化程度。观察抑制或增加BMP2信号对Decorin诱导的血管平滑肌细胞钙化和Msx2、Osterix表达的影响。【结果】Decorin能促进人血管平滑肌细胞钙化,同时上调BMP2、Msx2、Osterix的表达水平。抑制BMP2能减轻Decorin诱导的细胞钙化,而增加BMP2加速Decorin诱导的细胞钙化。【结论】Decorin通过调节BMP2促进人血管平滑肌细胞钙化。[Objective] The study aims to elucidate whether Decorin regulates calcification of vascular smooth muscle cells （VSMC） involving BMP2 signaling.[Methods] In vitro model of human VSMC calcification was used in this study.Decorin was overexpressed in cultured human VSMC using adenovirus-mediated gene transfer.The expression of BMP2,Msx2 and Osterix was analyzed by qRT-PCR and calcification was assessed by alizarin red staining.In addition,we checked the role of BMP2 signaling in decorin-induced vascular calcification by blocking or augmenting BMP2 signaling.[Results] We found that Decorin accelerated human VSMC calcification and up-regulated the expression level of BMP2、Msx2 and Osterix.Inhibition of BMP2 signaling attenuated Decorin-induced VSMC calcification,whereas augmentation of BMP2 signaling accelerated Decorin-induced VSMC calcification.[Conclusion] We demonstrate that Decorin induces human VSMC calcification via activation of BMP2 signaling.
