摘 要:目的探讨外源性转化生长因子β1(TGF-β1)抗体对SiO2刺激的小鼠肺成纤维细胞生长的抑制作用及细胞周期的影响。方法实验分为3组:空白对照,SiO2(100 mg/L)组,SiO2(100 mg/L)+TGF-β1(1.0μg/ml)抗体组。应用MTT法检测小鼠肺成纤维细胞增殖的变化,流式细胞仪检测细胞周期。结果各处理组细胞增殖差异有统计学意义(P<0.05),SiO2组细胞增殖最明显,其次为SiO2+TGF-β1抗体组;与空白对照组比较,SiO2组G0/G1期细胞百分率显著降低,S期及G2/M期细胞百分率增加,差异均有统计学意义(P<0.05),经TGF-β1抗体处理后,G0/G1期细胞百分率与SiO2组比较明显增加,S、G2/M期细胞百分率显著降低(P<0.05)。结论 TGF-β1抗体可能通过阻止SiO2刺激的小鼠肺成纤维细胞由G0/G1期进入S期,从而抑制肺成纤维细胞增殖。Objective The study aimed to explore the effect of exogenous transforming growth factor β1 (TGF-β1) antibody on the proliferation and cell cycle of SiO2-stimulated mice lung collagenoblasts. Methods The experimental mice was divided into 3 groups: a blank control group, a SiO2 (100 mg/L) group, and a SiO2 (100 mg/L) + TGF-β1 (1.0 μg/ml) antibody group. MTT assays and flow cytometry were used to measure the changes in proliferation and cell cycle of mice lung collagenoblasts, respectively. Resuits Differences in cell proliferation were statistically significant among the three groups (P〈0.05), with the SiO2 group exhibiting the fastest cell proliferation, followed by the SiO2+TGF-β1 antibody group. As compared with the blank control group, the SiO2 group showed a reduced percentage of G0/G1 cells and an increase in the percentage of S and GJM cells; the changes were statistically significant (P〈0.05). After treating with TGF-β1 antibodies, the percentage of G0/G1 cells increased and the percentage of S and G2/M cells decreased significantly, as compared with those of the SiO2 group (P〈0.05). Conclusion TGF-β1 antibodies may inhibit proliferation of lung collagenoblasts possibly by blocking SiO2-stimulated mice lung collagenoblasts from progressing into S phase from G0/G1 phase.
