摘  要:目的筛选曲古菌素A作用胃癌细胞系SGC-7901后明显表达下调的基因,检测其在临床胃癌组织中的表达水平。方法采用基因芯片技术,检测胃癌细胞系SGC-7901经过曲古菌素A干预后基因衷达的变化,筛选明显表达下凋的基因,应用免疫组化检测候选分子在胃癌及癌旁正常组织中的表达。结果胃癌细胞系SGC.7901经75ng/mL曲古菌素A作用48h后,Dickkopf related protein-1(DKK1)基因表达下调,差异具有统计学意义(P=0.0370)。免疫组化显示胃腺癌组织中DKK1基因表达明显高于周围正常胃组织,二者表达差异具有统计学意义(P=0.000)。结论DKK1基因在曲古菌素A作用胃癌细胞系SGC-7901后表达下调。DKK1基斟在胃腺癌组织中表达增高。Objective We investigated that the down-regulated genes were involved in Trichostatin A treatment in gastric cancer SGC-7901 cells. The expression of the candidate gene was detected in clinical gastricadenocarcinoma cases. Methods The changes of gene expression were detected in gastric cancer cell line SGC-7901 after Trichostatin A intervention by using gene chip technology. The down- regulated genes were screened and the expression was tested in gastric adenocarcinoma tissues by immunohistochemical analysis. Results Gastric cancer SGC-7901 cells with 75 ng/mL after 48 h of Triehostatin A treatment showed down-regulation of Dickkopf related protein-1 ( DKK 1 ) gene expression. The differences of DKKI expression were significant (P = 0. 037 0 ). Immunohistochemistry result showed that the expression of DKKI in gastric adenocarcinoma was significantly tissues (P =0. 000 ). Conclusion DKK1 expression was higher than that in the surrounding normal decreased in gastric cancer SGC-7901 cells 'after Trichostatin A treatment. DKK1 is overexpressed in gastric adenocarcinoma tissue.
