摘 要:近年我国头颈肿瘤发病率呈上升趋势,虽然医疗技术不断发展,但头颈肿瘤的五年生存率并无明显提高。选择性环氧化酶-2(cyclooxygenase-2,COX-2)抑制剂通过抑制前列腺素(prostaglandin,PG)的合成从而抑制肿瘤的发生与发展,是目前肿瘤研究的热点之一。本文旨在对有关选择性COX-2抑制剂的作用机制及其对头颈肿瘤预防效果的动物实验研究进行综述,为在此基础上深入开展临床研究提供借鉴。Attention should be paid to the situation of recently increased incidence of head and neck cancer. Despite advances in radiotherapy and chemotherapy, the survival of patients with head and neck squamous cell carcinoma has not improved significantly over the past three decades. Cyclooxygenase-2 (COX-2) , an enzyme that catalyzes the synthesis of prostaglandin, is being intensively evaluated as a potential molecular target for chemopreventive agents. Also, selective COX-2 inhibitors have become a hot topic to suppress the growth and metastasis of tumors. This article provides an overview of COX-2 biology, the effect through animal experiments and rationale for investigating whether selective COX-2 inhibitors will be useful in preventing or treating head and neck squamous cell carcinoma.
