摘 要:为探讨牛鞭草(Hemarthria spp.)品种间的遗传多样性,从86 对EST-SSR 引物中筛选出8 对引物对牛鞭草属6 个品种进行指纹图谱的构建及遗传多样性分析.结果表明,8 对EST-SSR 引物对牛鞭草属6 个品种共扩增出193 条清晰条带,多态性条带161 条,多态性比例为83.4%.每条引物的多态信息含量(PIC)为0.480 ~ 0.695,平均为0.602.UPGMA 聚类分析表明,牛鞭草属6 个品种在相似系数为0.652 处可分为两大类群.8 对EST-SSR 引物均能将6 个品种完全区分开,以3 对EST-SSR 引物扩增的电泳图谱为基础,建立了牛鞭草属6 个品种的指纹图谱标准模式图,每个品种都有唯-的指纹图谱.牛鞭草属6 个品种的平均Nei’s 基因多样性指数为0.333,平均Shannon 信息指数为0.496,品种间的相似系数介于0.399 ~ 0.782 之间.可见,牛鞭草属植物品种的遗传多样性较丰富,种间差异明显.In order to understand the genetic diversity of Hemarthria cultivars, the DNA fingerprints of sixHemarthia cultivars were constructed by using 8 pairs of EST-SSR primers screened from 86 pairs, and the geneticdiversities among them were analyzed. The results showed that eight pairs of ESR-SSR primers totally amplified194 bands, in which 161 bands were polymorphic bands, accounting for 83.4%. The polymorphic informationcontent (PIC) ranged from 0.480 to 0.695, with an average of 0.602. The UPGMA analysis showed that sixcultivars of Hemarthia could be divided into two groups at the similarity coefficient of 0.652, and 6 cultivarscould be completely separated by eight pairs of ESR-SSR primers. Based on electrophoretogram amplified by 3pairs primers, the standard DNA fingerprinting for 6 Hemarthia cultivars were constructed, and each cultivars hadan unique fingerprint. The average Nei’s genetic diversity and Shannon’s information index were 0.333 and 0.496,respectively. The similarity coefficient of 6 cultivars of Hemarthia ranged from 0.399 to 0.782. Therefore, it wassuggested that 6 cultivars of Hemarthia had rich genetic diversity and significant differences between species.
