摘  要:目的建立快速、灵敏的头孢氨苄残留荧光免疫层析试纸条检测法。方法以Eu-SiO2荧光纳米颗粒作为标记物,共价偶联于头孢氨苄单克隆抗体,将头孢氨苄-卵清蛋白全抗原和羊抗小鼠抗体喷涂于硝酸纤维素膜分别作为检测线和质控线,以竞争性免疫抑制反应模式制备了头孢氨苄残留荧光免疫层析试纸条。结果通过优化实验后制备的免疫层析试纸条对头孢氨苄残留的检测限为25 ng·mL-1,可在30 min内判读结果,与头孢菌素类、青霉素等的12种抗生素均无交叉反应,具有良好的特异性。结论本实验所制备的头孢氨苄残留荧光免疫层析试纸条具有操作简便、快速灵敏、便于普及等优点,为该类抗生素的检测提供了一种新型的检测方法。OBJECTIVE To develop a rapid and sensitive fluorescent immunochromatographic strip for the detection of cefalexin residue. METHODS The Eu-silica nanoparticles were synthesized by using RP-microemulsion technology. The Eu-silica nanoparti- cles conjugated with cefalexin monoclonal antibody was used as a fluorescent label. The fluorescent immunoehromatographic strip for the detection of cefalexin based on competitive inhibition immunoassay format was developed. The cefalexin-ovalbumin and goat anti-mouse IgG were immobilized on the nitrocellulose membrane as the test line and control line. RESULTS After experiment optimization, the developed strip could provide detection results within 30 rain and the limit of detection of cefalexin was 25 ng mL-l. There was no cross-reactivity with 12 kinds of antibiotics including cephalosporins and penicillin. CONCLUSION The fluorescence immunochroma- tographic strip is rapid, sensitive, and easy to operate, which may be a powerful tool to detect antibiotics in the future.
